摘 要:通过富集、分离和纯化等步骤,从活性污泥中分离筛选了两株能以亚硝酸盐为唯一氮源生长的异养硝化细菌53和N419.通过对其形态、生理生化、16 S rDNA基因序列、(G+C)mol%含量的测定及DNA-DNA杂交分析,初步鉴定53为施氏假单胞菌(Pseudomonas stutzeri),N419为芽孢杆菌属的一种(Bacillus sp.).并验证了这两株菌的硝化能力,结果表明,当液体培养及初始亚硝态氮浓度为2.03 g/L左右时,经振荡培养27 h,菌株53和N419对亚硝态氮的去除率分别达到87.9%和90.8%,总氮的去除率为54.7%和63.5%.Two strains of heterotrophic nitrifiers designated as 53 and N419,capable of utilizing nitrite as sole nitrogen source were isolated from the activated sludge through the procedure of enriching,separating and purifying.Based on their morphological and biochemical characters,16 S rDNA sequence,(G+C) mol% and DNA-DNA hybridization,strain 53 and N419 were identified as Pseudomonas stutzeri and Bacillus sp.,respectively.Results showed that when the initial nitrite-nitrogen concentration was 2.03 g/L,the nitritenitro-gen removal rates of 53 and N419 within 27 h were 87.9% and 90.8% respectively,and the removal rates of total nitrogen were 54.7% and 63.5% respectively.
