摘 要:[目的 ]揭示引流熊胆灌胃对小鼠肝脏谷胱甘肽S 转移酶及细胞色素P 45 0体系活力的影响及其与小鼠肝脏解毒能力的关系 .[方法 ]给昆明种小鼠灌胃引流熊胆水溶液后 ,分别在 1,4,8d测定了肝脏胞浆及微粒体中谷胱甘肽S 转移酶、细胞色素P 45 0体系的活力及蛋白质含量 ,并对 2 4h前灌胃引流熊胆水溶液的小鼠腹腔注射环磷酰胺 .[结果 ]引流熊胆水溶液可降低胞浆谷胱甘肽S 转移酶以 1 氯 2 ,4 二硝基苯为底物时的比活力 ,胞浆谷胱甘肽S 转移酶比活力由 (2 2 36± 312 )mmol/ (min·g)降至(10 0 4± 136 )mmol/ (min·g) .每 g组织活力由 (15 2 6 6± 1883)nmol/ (min·g)降至 (6 788± 10 13)nmol/ (min·g) ;微粒体谷胱甘肽S 转移酶比活力、细胞色素P 45 0活力、胞浆及微粒体蛋白质含量无显著变化 .受试小鼠在 7d内无 1只死亡 ,对照组小鼠 7d内死亡数为 8只 .[结论 ]引流熊胆可降低胞浆内谷胱甘肽S 转移酶的活力 ,但不影响微粒体内谷胱甘肽S 转移酶及细胞色素P 45 0的活力 ,并可增强小鼠对环磷酰胺的耐受能力 .OBJECTIVE To study the effects on activities of drug metabolizing enzymes in mice liver and its liver detoxicating ability by orally given bear bile.METHODS Male mice were given drainage bear bile orally.Samples were selected at 1st, 4th and 8th day for drug metabolizing enzyme's activities study.Glutathion s tranferase,cytochrome P 450 activity and protein content was measured by Habig, Omura Sato and Lourg's method.20 mice were abdominal injected with cyclophosphamide after drainage bear bile given orally.RESULTS Data observed shows that glutathion s tranferase activity toward CDNB in liver's cytosolic fractions has a decrease.Maxium decrease from (2?236±312)mmol/(min·g) to (1?004±136)mmol/(min·g) and (15?266± 1?883 )nmol/(min·g) to (6?788±1?013) nmol/(min·g) ,but in microsome fractions,neither the activities of glutathion s tranferase nor cytochrome P 450 are statistically changed.7 days survival rates after cyclophosphamide abdominal injection shows that with a decrease of hepatic cytosolic glutathione s transferase activity,bear bile group were higher(mice suvired:0/20 to 8/20).CONCLUSION Drainage bear bile can decrease the glutathione s transferase activity in liver's cytosolic but has no effect on microsome glutathione s transferase and cytochrome P 450,it can also decrease toxic effects caused by cyclophosphamide .
