摘 要:目的研究七氟醚在亚麻醉浓度(1.8%,相当于0.5 MAC)下对新生大鼠脑皮质凋亡和磷酸化JNK(c-Jun氨基末端激酶)1/2、ATP合成酶β亚基表达的影响。方法新生7 d SD大鼠6窝,每窝取10只,共60只,随机平均分配至七氟醚组和对照组。七氟醚组新生鼠吸入1.8%七氟醚4 h,对照组新生鼠暴露于空气4 h。在吸入七氟醚或空气结束时每窝每组各取1只幼鼠心脏穿刺抽血检测血糖和血气变化(n=6)。在麻醉恢复后2h和3 d,每窝每组各取1只幼鼠取新鲜脑皮质(n=6),Western blot检测各组磷酸化JNK1/2、ATP合成酶β亚基和Caspase-3的表达。结果 1.8%七氟醚麻醉4 h对幼鼠呼吸抑制轻微,血糖、血气分析结果与对照组比较差异均无统计学意义(P>0.05)。恢复2 h后,七氟醚组Caspase-3表达较对照组上升419.9%(P<0.05),ATP合成酶β亚基较对照组下降39.6%(P<0.01);3 d后,七氟醚组与对照组Caspase-3和ATP合成酶β亚基的表达比较差异无统计学意义(P>0.05)。七氟醚麻醉对磷酸化JNK1和JNK2的表达无明显影响。结论亚麻醉浓度七氟醚麻醉可增加新生7 d幼鼠脑皮层神经元凋亡,ATP合成酶β亚基表达下降可能是其诱导凋亡的机制之一。Objective To investigate the effect of sevoflurane under 0.5 MAC on cell apoptosis and expression of phosphorylated JNK1/2 and ATP synthase β subunit in new born rat cerebral cortex. Methods Sixty neonate rats (7 - day old) from 6 litters ( 10 pups from each litter) were randomly assigned into 4 groups. The rats were exposed to 1.8% sevoflurane or air as control for 4 h in sevoflurane groups and control groups, respectively. The blood sugar and blood gas assessments were carried out with cardiac puncture. After 2 - hour and 3 - day recoveries, the brain tissues were obtained for assessment of phosphorylated JNK1/2, ATP synthase β subnnit and Caspase - 3 using Western blot. Results Sevoflurane exposure did not affect blood sugar and ABG result in neonatal rats, but Caused significantly temporary Caspase - 3 elevation by 419.9% ( P 〈 0. 05 ) and reduction of ATP synthase β snbunit by 39. 6% ( P 〈 0. 05 ) after 2 h recovery, which were all reversed after 3 d recovery. There was no significant change of phosphorylated JNK1/2. Conclusion sevoflurane exposure induces temporary apoptosis in neonatal rat cortex, probably via depression of ATP synthase subunit.
