摘  要:利用简单序列重复区间(ISSR)分子标记技术,对广东广州和海南三亚2个地区云斑尖塘鳢群体的遗传多样性进行分析。采用加拿大哥伦比亚大学设计的ISSR引物,经过筛选后采用4条引物对云斑尖塘鳢的两个群体各12条进行ISSR分析,扩增出31个位点,两个群体的多态位点比例为83.87%,Shannon信息指数0.4591,群体间遗传距离为0.1350,遗传分化系数0.1493,基因流2.8499,说明云斑尖塘鳢群体间分化主要受到群体内部的变异的影响,而不是遗传漂变,广州地区的云斑尖塘鳢群体遗传多样性优于三亚群体。UPGMA聚类分析结果表明,云斑尖塘鳢没有群体间界限。Inter simple sequence repeat (ISSR) were applied to research the genetic diversity of two populations of Oxyeleotris marmoratus Bleeker. They were collected from Guangzhou (Guangdong) and Sanya (Hainan). Through the PCR amplification, four primers (834, 855, 859, 895) were chosen from UBC primer set #9 (Microsatellite). The primers were amplified 31 reproducible DNA fragment in the two populations. The percentage of polymorphic were 83.87%. Shannon's information index was 0.4591 among two populations. The average genetic distance was 0.1350. Gst was 0.1493 and Nm was 2.8499. The results showed that differentiation among populations was mainly influenced by the variability within groups, rather than the genetic drift. The genetic diversity of Guangzhou population was superior to Sanya. The two populations didn't have obvious group boundaries by the UPGMA tree based on ISSR data.
