摘  要:为了进一步提高猪精液的冻后品质,为中药提取物在猪精液冷冻中的应用提供参考数据,采用手握法采集成年种公猪精液,预处理后用添加不同剂量盐酸川穹嗪(0、0.08、0.10、0.12 mmol/L)和葛根素(0、0.001、0.01、0.10 mmol/L)的稀释液稀释,经平衡、冷冻、解冻后采用考马斯亮蓝对精子进行染色,然后在显微镜下检查精子畸形率和顶体完整率。结果表明,不同浓度的盐酸川穹嗪均能显著降低冻后精子的畸形率,显著提高冻后精子的顶体完整率,其中0.12 mmol/L盐酸川穹嗪效果最好。一定量葛根素不能显著降低冻后精子的畸形率,也不能显著提高冻后精子的顶体完整率。The objective of the present study was to further improve the quality of frozen-thawed boar semen and provide the references for the application of the two Chinese medicine extracts to boar semen cryopreservation. Samples were collected by the glovedhand technique from boars of proven fertility. After a preprocessing period, semen sample was extended in the diluents containing ligustrazine hydrochloride (0, 0.08, 0.10, 0.12 mmol/L) or puerarin (0, 0.001, 0.01, 0.10 mmol/L) and equilibrated. Finally frozen-thawed boar spermatozoa were stained with Coomassie brilliant blue to detect their abnormality and acrosomal integrity using a light microscope. The results showed that different concentrations of ligustrazine hydrochloride added to boar semen freezing extenders, of which 0.12 mmol/L of ligustrazine hydrochloride was the best, significantly reduced the abnormality or increased the aerosomal integrity of frozen-thawed spermatozoa. Addition of a certain amount of puerarin to freezing diluents neither significantly reduced the abnormality nor advanced the acrosomal integrity of frozen-thawed boar spermatozoa.
