CiaRH生物信息学分析及其在不同猪链球菌血清型中的分布

摘  要:目的对猪链球菌2型中二元信号转导系统CiaRH的序列进行分析并评估该系统在不同血清型中的分布。方法利用生物信息学方法分析其组分蛋白的结构域。PCR及序列分析手段评估CiaRH在不同血清型猪链球菌中的分布情况。结果比对结果显示,CiaRH同源蛋白存在于链球菌属的不同种细菌中。结构分析发现,在CiaH的N-端包含两个跨膜区,其间的间隔区域发现一个可能用来结合环境信号的PDC结构域;CiaH的C-端为保守的HisKA及HATPase_C结构域。PCR及序列比对结果显示,CiaRH在不同血清型猪链球菌中广泛分布。结论提示CiaH为胞外感应型组氨酸激酶。在CiaR的N-端为保守的信号接收结构域REC,而C-端为具有DNA结合功能的trans_reg_C结构域。This study aimed to analyze the sequences of a two-component signal transduction system(TCSTS) CiaRH from S.suis 2 and evaluate the distribution of this TCSTS on different serotypes.The BLAST results showed that the CiaRH orthologs were found in different species of the genus Streptococcus.Structure analysis on CiaRH revealed that two transmembrane regions existed at the N-terminal of CiaH,between which a PDC domain that would anticipate in signals binding was predicted;the highly conserved domains HisKA and HATPase_C were found at the C-terminal of the protein.These results suggest that the CiaH belongs to the periplasmic(or extracellular)-sensing HKs group.The conserved REC domain that involved in signal receiving and the DNA binding domain trans_reg_C were found located at the N-terminal and C-terminal of CiaR,respectively.Combined results of PCR detection and BLAST sequence suggest that the CiaRH are widely distributed in different serotypes of S.suis.
