葛根素与雌二醇对去卵巢大鼠子宫的影响

摘　　要：目的观察长期联合使用葛根素、雌二醇对去卵巢大鼠子宫组织结构的影响。方法将40只健康雌性5月龄SD大鼠随机分为5组(n=8):假手术组、去卵巢模型组、葛根素组、雌二醇组、葛根素-雌二醇组。假手术组大鼠只切除其卵巢旁的一小块脂肪垫;其余4组建立去卵巢大鼠骨质疏松症模型。1周后,葛根素组大鼠皮下注射葛根素50mg/kg,1次/d;雌二醇组大鼠皮下注射雌二醇200μg/kg,2次/周;葛根素-雌二醇组大鼠皮下注射葛根素(25mg/kg,1次/d)的同时还皮下注射雌二醇(100μg/kg,2次/周)。药物干预20周后取各组大鼠血清和子宫组织,观察血清雌二醇水平和子宫组织的形态学表现。结果雌二醇组大鼠血清雌二醇水平、子宫管径、子宫内膜面积、肌层面积和子宫腺面积明显高于去卵巢模型组、葛根素组及葛根素-雌二醇组(P<0.05)。葛根素-雌二醇组与葛根素组比较,前者大鼠血清雌二醇水平、子宫管径、子宫内膜面积、肌层面积和子宫腺面积有增加趋势,但差异无统计学意义(P>0.05)。结论小剂量雌二醇与葛根素联合或较大剂量葛根素对子宫的促增生作用较弱。Objective To observe the effect of long term combination therapy with puerarin and estradiol on uterine structure of ovariectomized rat. Methods Forty healthy 5-month-old female rats were randomly divided into 5 groups（n= 8） ：sham-operated, ovariectomized, puerarin,estradiol and puerarin-estradioI group. Rats in sham-operated group were subjected to removal of ovarian fat pads only,and ovariectomized-induced osteoporosis rats model were established in the rest 4 groups. One week after modeling, rats in the puerarin group were subcutaneously injected with 50 mg/kg puerarin once daily, and rats in the estradiol group with 200 ug/kg estradiol twice weekly,and rats in puerarin-estradiol group with puerarin（25 mg/kg, once daily） and estradiol（100 /~g/ kg,twice weekly） simultaneously. After 20 weeks of drug intervention, serums and uterine tissues of rats in each group were col- lected,and the level of serum estradiol and morphological structure of uterus were observed. Results Serum estradiol level,uterine diameter, endometrium area,myometrium area and uterine gland area of rats in estradiol group were significantly higher than those in ovariectomized, puerarin and puerarin-estradiol group（P〈0. 05）. Compared with puerarin estradiol group and puerarin group, rats in the former showed a increasing trend of serum estradiol level, uterine diameter, endometrium area, myometrium area and u terine gland area,with no statistically significant（P〉0.05）. Conclusion Both low dose of estradiol and puerarin combination and relatively high dose of peuerarin possess small effect on uterine proliferation.
