摘 要:目的运用代谢组学方法研究角叉菜胶致大鼠足肿胀后的变化及双黄连口服液发挥抗炎作用的相关代谢途径。方法大鼠血清和尿液的代谢指纹利用核磁共振氢谱(1H-NMR)进行表征,综合使用多变量统计分析方法如主成分分析、正交偏最小二乘判别分析和单变量统计分析方法探讨炎症引起相关代谢标志物含有量变化。结果双黄连口服液可以使角叉菜胶致大鼠足肿胀后发生紊乱的机体代谢表型回归到正常水平。在血清和尿液中分别选取了12种和10种生物标志物含有量变化作对比。结论双黄连口服液能有效地缓解角叉菜胶导致的大鼠机体脂质和糖代谢失衡,同时,可以调节肠道菌群。AIM To investigate the metabolic profile of carrageenan-induced hind paw oedema in rats and the intervention of Shuanghuanglian Oral Liquid. METHODS Proton nuclear magnetic resonance ( 1 H NMR) was used to analyze the rat serum and urine metabolic profiles. Multivariate data analysis methods (orthogonal partial least squares discriminant analysis, principal components analysis) and univariate statistical analysis methods were used to analyze the metabolome difference between the control and carrageenan treated samples, and to identify the relevant metabolic biomarkers. RESULTS After carrageenan treatment, the metabolic phenotype of rats greatly changed and Shuanghuanglian Oral Liquid had effective intervention. Twelve and ten potential biomarkers in rat serum and urine were selected to distinguish the changes in the markers before and after the treatment. CONCLUSION Shuanghuanglian Oral Liquid can effectively alleviate the disturbance of lipids and glucose metabolism and modulate the gut mieroflora environment in rats.
