摘  要:目的比较大蒜活性物质对高血脂小鼠血脂代谢的影响。方法血浆中血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)和丙二醛(MDA)的水平测定以及超氧化物歧化酶(SOD)活力测定均采用试剂盒方法测定;3-羟-3-甲戊二酸单酰辅酶A还原酶(HMG-CoA还原酶)活力通过NADPH光吸收的减少量进行测定。结果大蒜素和蒜氨酸+蒜氨酸酶能降低高血脂小鼠TC、LDL-C和MDA的水平,抑制HMG-CoA还原酶活力,提高HDL-C水平和SOD酶活力。与高血脂组相比,处理各组有显著差异,并呈现剂量依赖性。结论大蒜素和蒜氨酸+蒜氨酸酶都具有良好的降血脂作用,其机制可能与阻断脂质过氧化、抑制胆固醇合成途径中的限速酶———HMG-CoA还原酶的活性密切相关,并且蒜氨酸+蒜氨酸酶降低TC水平和HMG-CoA还原酶活性、提高HDL-C水平和SOD酶活力的效果优于大蒜素。AIM To compare the effects of garlic activity substances on plasma-lipids metabolism in mice fed high fats diet. METHODS Plasma total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), malondialdehyde (MDA) and superoxide dismutase (SOD) activity were assayed by their kits. 3-hydroxy-3methylglutaryl-CoA (HMG-CoA) reductase activity was measured by the decrement of NADPH absorption. RESULTS Allicin and the mixture of alliinase and alliin could decrease the level of TC, LDL-C, MDA and I-IMG-CoA reductase activity, while increasing HDL-C and SOD activity in mice fed high fats diet. Compared with the hyperlipidemia group, there were significant differences among each treatment group with dose-dependent relationship. CONCLUSION Allicin and the mixture of alliinase and alliin whose mechanism may closely relate to block lipid peroxidation and inhibit the activity of rate-limiting enzyme of cholesterol synthesis--HMG-CoA reductase activity all have a better lipid-lowering effect. The effect of mixture of alliinase and alliin is better than that of allicin on reducing triglycerides, HMG-CoA reductase activity and increasing HDL-C and SOD activity.
