摘 要:目的建立同时测定Caco-2细胞模型中黄芩苷、汉黄芩苷、汉黄芩素和千层纸素A的HPLC法,探讨黄芩总黄酮在Caco-2细胞上的吸收机制。方法利用人源结肠腺癌细胞系Caco-2细胞单层模型研究黄芩苷、汉黄芩苷、汉黄芩素和千层纸素A由肠腔侧(AP侧)到基底侧(BL侧)和BL侧到AP侧两个方向的转运过程。应用HPLC-UV对上述4个黄酮成分进行分析,计算转运参数和表观渗透系数(Papp)。结果黄芩总黄酮给药后,黄芩苷、汉黄芩苷、汉黄芩素和千层纸素A由AP→BL侧的Papp分别为(1.65±0.38)×10-6、(1.58±0.21)×10-6、(1.57±0.15)×10-6和(5.74±0.18)×10-6cm/s,由BL→AP侧的Papp分别为(1.23±0.24)×10-6、(1.22±0.16)×10-6、(1.27±0.07)和(5.13±0.19)×10-6cm/s;表观渗透率(PDR)分别为0.75、0.77、0.80和0.89。结论 4个黄酮类主要以被动吸收方式进入体内。AIM To establish an RP-HPLC method for simultaneously determining the total flavonoids from Scutellariae Radix including baicalin, wogonoside, wogonin and oroxylin A, and to investigate their absorption characteristic in human intestine after being administrated. METHODS By using intestinal epithelial cell Caco-2 model, the permeability of baicalin, wogonoside, wogonin and oroxylin A were studied from apical side ( AP side) to basolateral side ( BL side) or from BL side to AP side. The four flavonoids were measured by RP-HPLC with UV detector. Transport parameters and apparent permeability coefficients (Papp) were then calculated. RESULTS The Papp values of baicalin, wogonoside, wogonin and oroxylin A were (1.65 ± 0.38) × 10^-6, (1.58 ± 0.21 ) × 10^-6, (1.57±0.15) × 10^-6 and (5.74±0.18) × 10^-6cm/s from AP side to BL side, and (1.23±0.24) × 10^-6, ( 1.22 ±0. 16) × 10^-6, ( 1.27 ±0. 07) and (5.13 ±0. 19) × 10^-6 cm/s from BL side to AP side, respectively. The permeability direction ratios (PDR) of baicalin, wogonoside, wogonin and oroxylin A were 0. 75, 0. 77, 0. 80 and 0. 89, respectively. CONCLUSION Baicalin, wogonoside, wogonin and oroxylin A in total flavonoids from Scutellariae Radix could be absorbed across intestinal epithelial cells in passive absorption pattern forms.
