中医“血瘀、阳亢、痰浊”高血压动物模型构建的研究

摘　　要：目的:构建一种具备中医"血瘀-阳亢-痰浊"证候特点的高血压大鼠模型,为相关研究提供载体。方法:将24只5周龄雄性自发性高血压大鼠(SHR)随机分为模型组(SHR-M)及对照组(SHR-C),同时设置9只年龄和性别相匹配的WKY大鼠(WKY-C)对照。其中SHR-C及WKY-C组始终以普通大鼠饲料喂养,而SHR-M组则接受持续高脂饮食喂养并且前4周以附子汤灌胃。12周后观察各组大鼠的血压、易激惹程度,并通过腹主动脉取血测定AngⅡ含量,空腹血糖、胰岛素、甘油三酯及胆固醇水平、血黏度等指标。结果:12周后SHR-M组大鼠血压高于其他两组大鼠(P<0.05),SHR-M组大鼠具有更高的易激惹程度(P<0.01),SHR-M组大鼠的血浆AngⅡ、甘油三酯、胆固醇、血黏度等指标均显著高于其余两组(P<0.05),SHR-M组大鼠的HOMA-IR胰岛素抵抗指数显著高于其余两组(P<0.05)。结论:以SHR为基础持续高脂饮食喂养配合附子汤灌胃可成功建立具备中医"血瘀-阳亢-痰浊"证候特点的高血压大鼠模型。Objective：To establish a hypertensive rat model with blood stasis-liver Yang rising and turbid phlegm syndrome of traditional Chinese medicine. Methods ： 24 male SHRs, 5 weeks old, were randomly divided into two groups ： SHR model group （SHR-M） and SHR control group（SHR-C）. 9 sex and age matched Wistar-Kyoto rats （WKY-C）were served as another control group. The rats of SHR-C and WKY-C were fed with ordinary forage. The rats of SHR-M were fed with high fat diet and they were fed with monkshood at the first 4 weeks. After 12 weeks ,the blood pressure of the rats were measured and the irritability degrees of the rats were observed. The serum Ang lI , fasting blood-glucose, insulin, cholesterol, triglyceride and blood viscosity of all the rats were examined. Results： After 12 weeks,the rats of SHR-M had the highest blood pressure among the three groups （P 〈 0. 05）. The rats of SHR-M were more irritable than the others（P 〈 0. 01 ）. The above biochemical indicators of the SHR-M~ rats were significantly higher than the others（ P 〈 0. 05 ）. The insulin resistance index（HOMA-IR） of the SHR-M~ rats were also high- er than the others （P 〈 0. 05 ）. Conclusion：The hypertensive rat model with blood stasis-liver Yang rising and turbid phlegm syn- drome of traditional Chinese medicine can be established by the SHRs fed with high fat diet and monkshood.
