摘 要:目的:研究葛根素(Pue)对早期糖尿病肾病(DN)大鼠肾组织病理变化与骨形态发生蛋白-7(BMP-7)及mRNA表达的影响。方法:以链脲佐菌素(STZ)腹腔注射复制早期DN大鼠模型。动物分为正常对照、模型、依那普利(1mg/kg)与Pue高、中、低剂量(200、100、50mg/kg)组,灌胃给药,每天1次,连续5周。末次给药后检测大鼠24h尿量、体质量、肾/体质量比值(KW/BW);生化指标检测空腹血糖(FBG)、24h尿量尿素氮(BUN)、血肌酐(Scr)、胰岛素(Ins)、24h尿蛋白(Upro)及24h尿微量白蛋白(UmAlb);光镜和电镜观察肾组织形态学和超微结构的改变;免疫组化法和逆转录聚合酶链反应(RT-PCR)检测肾组织BMP-7蛋白及mRNA的表达。结果:与模型组比较,Pue高、中、低剂量组大鼠FBG、KW/BW、BUN、Scr水平降低,体质量、Ins水平升高,BMP-7蛋白及mRNA表达增多(P<0.01或P<0.05),肾组织病理形态学异常与超微结构病变改善;Pue高、中剂量组24h尿量、24hUpro、24hUmAlb显著降低(P<0.01或P<0.05)。结论:Pue可显著升高早期DN大鼠肾组织BMP-7蛋白、mRNA表达,改善早期DN大鼠肾功能病变,延缓DN发展。OBJECTIVE:To explore the effects of puerarin on the pathological change and protein and mRNA expressions of bone morphogenetic protein-7(BMP-7) in the kidney of rats with early diabetic nephropathy(DN).METHODS:Early DN rat model was induced by streptozotocin(STZ).The rats were divided into normal control group,model group,enalapril group(1 mg/kg)and puerarin high-dose,medium-dose and low-dose groups(200,100,50 mg/kg).They were given medicines intragastrically once a day for consecutive 5 weeks.After the last administration,the urinary volume,body weight,24 h urine volume,kidney weight/body weight,FBG,BUN,Scr,insulin(Ins),24 h urinary protein(Upro)and 24 h urinary microalbumin(UmAlb)were tested;the pathological changes and ultrastructures in renal tissue of DN rats were observed by optical and electron micrograph.Immunohistochemistry and RT-PCR were used to detect the protein and mRNA expressions of BMP-7 in the renal tissue of rats.RESULTS:Compared with model,FBG,KW/BW,BUN,Scr of puerarin high-dose,medium-dose and low-dose groups decreased while BW,Ins level and protein and mRNA expression of BMP-7 increased;the pathological changes and ultrastructureal lesion in renal tissue of DN rats were improved(P〈0.01 or P〈0.05).24 h urine volume,24 h Upro,24 h UmAlb in Pue high-dose,medium-dose groups decreased(P〈0.01 or P〈0.05).CONCLUSION:Puerarin can obviously improve renal function and increase the protein and mRNA expression of BMP-7 in kidney tissue,and delay the progression of DN in DN model rats.
