高脂加酒精灌胃法诱导大鼠脂肪肝成模及思考

摘　　要：目的探索简便易行的酒精性脂肪肝造模方法及成模时间,为保证酒精肝成模的稳定性和可靠性提供参考。方法30只SD大鼠随机分为2组。实验组给予40%、45%、50%梯度浓度酒精灌胃,正常对照组给予等量生理盐水代替酒精灌胃,于实验的2、4、5、8、12周末分批采血测定生化指标,后脱颈处死,取出肝脏石蜡包埋,HE染色后行病理学观察。结果实验组大鼠于第3周起观察到酒精性肝损伤的早期变化(如脂肪变性,点灶状坏死,炎性浸润等),并随时间变化呈逐渐加重的趋势。结论应用高脂加酒精灌胃法可成功复制大鼠酒精性脂肪肝模型。Objective To research a simple accurate induction method of alcoholic fatty liver model（AFL） in rats and modeling time,and provide reference for guaranting the stability and reliability of method of making AFL model.Methods 30 SD rats were divided into 2 groups randomly.Rats in the experiment group were given different gradient concentration（40%、45% and 50%） of alcohol by daily gavage,and rats in normal controls were given equivalent normal saline.All of the rats were randomly sacrificed at the end of 2nd,4th,5th,8th,and 12th weeks after experiment,blood samples were collected to measure the concentrations of blood proteins and enzymes.Paraffin-embedded liver tissues were stained by HE,and for pathological observation.Results The early changes of alcoholic hepatic injury in the experiment group,such as fatty degeneration,dot or piece necrosis and inflammatory infiltration,were observed in the 3rd week,and aggravated gradually along with the prolongation of time.Conclusion The model of AFL in rats can be successfully established by means of high-fat diet and gastric perfusion of alcohol.
