摘  要:目的探讨黄芩提取物对耐药性金黄色葡萄球菌的耐药抑制机理,为中药耐药抑制剂的研究开发提供一定的基础。方法采用SDS-PAGE电泳法测定黄芩提取物处理后耐药性金黄色葡萄球菌蛋白质电泳图谱变化情况;采用荧光分光光度法测定菌体内环丙沙星药物的蓄积量。采用半自动生化分析仪测定细菌细胞外液中有关酶活性。结果耐药金黄色葡萄球菌在分子量为43kDa~55 kDa范围内有蛋白质条带的缺失或表达量的降低,经黄芩提取物处理后这些蛋白质可恢复表达或表达量增加;黄芩提取物可使菌体内环丙沙星蓄积浓度明显增加,但对细菌细胞膜通透性无明显影响。结论黄芩提取物具有恢复耐药菌某些蛋白质的表达和抑制膜上外排泵的作用,因此具有明显的耐药抑制作用。Objective To probe the resistance inhibition mechanism of Baical Skullcap root extracts to Staphylococcus aureus and to provide theoretical basis for development of resistance inhibitor.Methods The accumulated concentration of Ciprofloxacin was determined by fluorospectrophotometry and the outer-membrane protein(OMP) composition was analyzed by SDS-PAGE electrophoresis after Baical Skullcap root extract was treated,enzyme activity in bacterial extracellular fluid was detected by semi-automatic biochemical analyzer.Results The accumulated concentration of medicine Ciprofloxacin within the experiment strains treated with coptis extract was obviously higher than control strains(P0.01);In resistance strain,the relative content of OMP(the relative content of OMP of the resistance strain) from 43kDa to 55kDa was less or lacked compared with Staphylococcus aureus ATCC25923 strain.The amount that lack of 43kDa to 55 kDa membrane proteins may be rebound expressed after Baical Skullcap root extract treated,however Skullcap root extract had no obvious effect to bacterial cell membrane permeability.Conclusion Baical Skullcap root extract can recover and increase membrane protein expression,and excretion pump can be inhibited by Baical Skullcap root extract,it was plausible that Baical Skullcap root extract has resistance inhibition effect to resistance Staphylococcus aureus.
