摘  要:目的测定梗阻性黄疸大鼠小肠组织IgA+、TGF-β+及IL-8 mRNA+细胞数及门静脉血内毒素含量,探讨梗阻性黄疸时损害肠黏膜屏障功能进而引起肠源性内毒素血症的病理生理机制。方法用Wistar雄性大鼠48只,分为假手术组及梗阻性黄疸组,并于术后4、7和14 d分别测定大鼠门静脉血内毒素及结合胆红素含量,取小肠组织测定IgA+、TGF-β+及IL-8 mRNA+细胞数。结果梗阻性黄疸组与假手术组相比较内毒素含量明显增加,梗阻性黄疸组内比较随梗阻时间延长内毒素浓度增加更加明显。梗阻性黄疸组与假手术组相比较,IgA+、TGF-β+及IL-8 mRNA+细胞数明显减少。梗阻性黄疸组组内相比,IgA+、TGF-β+及IL-8 mRNA+细胞数随胆道梗阻时间延长而减少。结论梗阻性黄疸致小肠黏膜固有层内IgA、TGF-β及IL-8 mRNA的表达减少,使之成为削弱肠黏膜屏障功能的原因之一,可能是导致和加重肠源性内毒素血症的一个重要原因。【Objective】 To study the mechanism of intestinal mucosal barrier damaging by the obstructive jaundice.【Methods】 48 male Wistar rats were randomly assigned into two groups,the same operation group(SO group) and the obstructive jaundice group(OJ group).Determine the endotoxin and DBIL of blood in the portal vein,determine the IgA+,TGF-β+ and IL-8 mRNA+ cell number on 4 d,7 d,14 d after the operation.【Results】 The concentration of endotoxin in the OJ group were higher than the other group,and inner the OJ group,the concentration of endotoxin were gradually enhanced along with the time.The IgA+,TGF-β+ and IL-8 mRNA+ cell number in OJ group were decreased than the other group,and inner the OJ group,the IgA+,TGF-β+ and IL-8 mRNA+ cell number were gradually decreased along with the time.【Conclusion】 The expression of the IgA,TGF-β and IL-8 mRNA in intestine were decreased in obstructive jaundice,and it probably cause the intestinal mucosal barrier damaging,and it would also be the one of the causation for endotoxemia.
