摘 要:参考GenBank上发表的猪星状病毒亚基因组序列,设计1对引物,对猪星状病毒广西流行株衣壳蛋白ORF2基因进行扩增,最终克隆得到PAstV/NNJG7株的衣壳蛋白完整序列。序列分析发现,其ORF2全长2 358bp,编码786个氨基酸;与猪星状病毒Ⅰ型参考株核苷酸同源性最高,为77.3%~79.4%;在ORF1b和ORF2连接处有一段高度保守的序列,在ORF2 3′端有一段长83bp非翻译序列及一个含43bp高度保守的茎环样基序。本试验不仅丰富了猪星状病毒衣壳蛋白基因序列,也为基因疫苗和诊断试剂盒的研制提供材料及理论依据。According to the subgenomic sequence of porcine astrovirus in GenBank, one pair of primers on porcine astrovirus capsid protein gene were devised and the current epidemic strain of Guangxi was am- plified. The complete sequence of PAstV/NNJG7 capsid protein was cloned. Sequence analysis revealed that the ORF2 contained 2358bp, encoding 786 amino acids. The nucleotide homologies were 77.3% to 79.4% compared with PAstV-I type reference strains. There was a highly conserved sequence in the junction between ORF2 and ORF1b. There was'a non-translated sequence with 83 nucleotides in length and a highly conserved stem-loop-like motif with 43 nucleotide in the 3' end of ORF2. This study not only enriched the capsid protein gene sequences of porcine astrovirus, but also provided materials and theoretical basis for the development of genetic vaccines and diagnostic kit of porcine astrovirus.
