摘 要:目的观察吴萸乙醇提取物(EEEO)对四氯化碳(CCl4)诱导大鼠急性肝坏死的影响。方法将Wistar大鼠分为5组[对照组、CCl4组、EEEO150mg/kg+CC14组、EEEO300kg/kg+CC14组和水飞蓟素(Silmyarin)50ms/kg+CC14组],分别用生理盐水、EEEO和Silmyarin灌胃5d,最后1d腹腔注射CCl4,15h后检测血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST);肝组织苏木素一伊红(HE)染色观察病变程度;分别用黄嘌呤氧化酶法和硫代巴比妥酸法检测肝匀浆中超氧化物岐化酶(SOD)、过氧化氢酶(CAT)和丙二醛(MDA)。结果CCl4组血清中ALT、AST的含量明显高于对照组[(345.4±51.6)U/ml、(621.7±143.5)U/m1VS(41.1±2.2)U/ml、(85.2±22.2)U/ml,P〈0.05],HE染色肝组织变性坏死累及全小叶。EEEO+CC14组血清中低剂量组ALT[(308.1±44.6)U/m1]与AST[(546.4±131.6)U/m1]含量和高剂量组ALT[(210.65-34.5)U/m1]与AST[(379.3±112.3)U/m1]的含量明显低于CCl4组(P〈0.05)HE染色病变程度较轻;胞浆中SOD和CAT活性提高、MDA水平降低(P〈0.05)。结论EEEO能显著减轻CCl4诱导的肝损伤,可能与EEEO抗脂质过氧化作用有关。Objective To investigate the protective effect of ethanol extract from Evodia officinalis Dode (EEEO) in a rat model of acute hepatic necrosis induced by carbon tetrachloride ( CC14 ). Methods Wistar rats were divided into four groups ( control, CC14, EEEO + CC14, and Silmyarin + CC14) , the four groups were given intragastrically with normal saline, EEEO for 5 d, respectively. In the last one day, these groups except for control group were injected peritoneally with CC14. Serum levels of alanine amin- otransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) were detected by au- tomatic biochemistry analyzer. Pathological changes of hepatic tissues were assessed by hematoxylin-eosine (HE) staining. The levels of superoxide dismutase ( SOD), catalase (CAT) and malondialdehyde (MDA) in liver homogenate were analyzed using xanthinoxidase and thio-barbituric acid, respectively. Results Compared the ALT [ (345.4 ± 51.6)U/ml ] and AST [ (621.7 ± 143.5 ) U/ml) ] of CC14 group with ALT [ (41.1 ± 2. 2) U/ml ] and AST [ ( 85.2 ± 22.2) U/ml ] of control group, the serum levels of ALT and AST in the CC14 group were increased significantly ( P 〈 0. 05 ). HE staining of liver tissue, the degeneration and necrosis were implicated to the whole hepatic lobules in the CC14 group. In EEEO + CC14 group, com- pared the ALT [ (308. 1 ±44. 6) U/ml] and AST [ (546. 4 ± 131.6) U/ml] of low dose EEEO + CC14 group with the ALT [ (210.6 ± 34. 5 ) U/ml ] and AST [ (379. 3 ± 112. 3 ) U/ml ] of high dose EEEO + CC14 group the serum levels of ALT and AST were decreased significantly in low dose EEEO + CC14 group ( P 〈 0. 05). The denaturation and necrosis of hepatic lobules, the level of SOD, CAT were increased and MDA decreased ( P 〈 0. 05 ) inendochylema. Conclusions EEEO can significantly relieve the CC14-induced hepatonecrosis. The role may be related to anti-lipid peroxidation.
