摘 要:利用生物信息学预测绵羊肺腺瘤病毒(OPAV)NM株囊膜蛋白优势抗原表位,为寻找用于绵羊肺腺瘤病诊断和预防的候选抗原表位提供参考。参照OPAV-NM株env基因序列(登录号:JQ837489),应用Gamier-Robson方法、SOPMA方法和网络服务器PSIPRED、Predictprotein预测OPAV-NM株囊膜蛋白的二级结构。用Kyte-Doolittle方法预测蛋白质的疏水区和亲水区,用Emini方法预测蛋白质表面可能性,以Jamson-Wolf方法预测蛋白的抗原指数,利用Bcepred、IEDB、ABCpred网络服务器预测囊膜蛋白的抗原性,然后综合评价囊膜蛋白的B细胞抗原表位。结果表明,囊膜蛋白(ENV)二级结构丰富,α螺旋和β折叠所占比例相对较多,也具有多处转角及无规则卷曲区域,提示OPAV-NM囊膜蛋白具有较多的优势抗原表位区段。这些优势抗原表位分别为第52~76,101~110,118~165,182~196,201~215,281~305,309~332,335~356,461~476,530~574位氨基酸,其中第461~476位和第530~574位的部分氨基酸序列位于胞外区,且第530~574区段位于en-OPAV和ex-OPAV的囊膜氨基酸序列的差异区,该段氨基酸序列对绵羊肺腺瘤病毒疫苗和诊断方法的研究有重要意义。The epitope of envelope protein of OPAV-NM strain was predicted by bioinformatics to find a better target site of antigens and can be used as candidate antigen in the diagnosis and precaution of ovine pulmonary adenomatosis. The secondary structure of envelope protein was predicted with the methods of Garnier-Robson, SOPMA and PSIPRED, Predictprotein network server based on the envelope protein of OPAV-NM strain genome which was submited to the GenBank:by our lab and the accession number is (JQ837489). Hydrophilicity, surface probility and antigenic index were obtained by the methods of Kyte- Doolittle, Emini and Jameson-Wolf respectively, and the antigenicity for the envelope protein was predic- ted by Bcepred, IEDB, ABCpred network server, Combining the results according to these methods, the B cell epitopes for envelope protein of OPAV-NM strain were predicted. The results showed that envelope protein of OPAV-NM strain has rich secondary structure, a helix regions and 13 sheet regions are more than 13 turn regions and coil regions; The results also showed that it has many 13 turn regions and coil regions, meaning the envelope protein of OPAV-NM strain has many epitopes, the predominant epitopes of the B cell may be the amino acid of the No. 52-76, 101-110, 118-165, 182-196, 201-215, 281-305, 309-332, 335- 356, 461-476 and 530-574, and the amino acid of the No. 461-476 and 530-574 are located in the extracellu- lar domain, and the amino acid of the No. 530-574 are located in the disparity region of envelope amino acid sequence of en-OPAV and ex-OPAV that has great significance in the vaccinology and diagnosis methods of OPAV.
