摘　　要：目的探讨CD4+T细胞在天疱疮致病性抗体产生中的作用。方法实验方面采用真核表达系统表达重组的天疱疮抗原桥粒芯糖蛋白3(Dsg3)胞外段功能区,联合不同佐剂(CFA或Alum)免疫C57BL/6小鼠,用免疫磁珠细胞分选(MACS)法分选出免疫后小鼠的CD4+T细胞,过继转移到T细胞受体β链(T cell receptor beta chain,TCRβ)缺陷小鼠,于转移后1、2、3、4周检测TCRβ缺陷小鼠体内CD4+T细胞的恢复,B细胞的增殖、活化,及特异性抗体的产生。结果过继转移后的TCRβ缺陷小鼠外周血中有CD4+T细胞的出现(比例有5%左右);CD4+T细胞进入缺陷小鼠脾脏中的淋巴小结,和B细胞相互作用;同时该小鼠血清中出现了特异性抗Dsg3抗体。结论 CD4+T细胞参与天疱疮致病性抗体的产生,在疾病中发挥极其重要的作用。Objective To investigate the role of CD4^＋T cells in producing the specific-autoantibodies in pemphigus. Methods We immunized the mice with purified rDsg3 emulsified with CFA or Alum. After immunization, CD4^＋T cells were separated from whole splenocytes by MACS. Following the adoptive transfer of these CD4^＋T cells to TCRβ deficient mice,the recovery of CD4^＋ T cell, and the production of specific antibodies were analyzed in the TCRβ deficient mice. Results After adotive transferred to the TCRβ deficient mice,5% of these CD4~T cells were detected in the peripheralblood of recipient mice. CD4^＋T cells showed up in the spleen lymphoid nodules of mice. Meanwhile the anti-Dsg3 antibodies were produced in these recipient mice. Conclusion These results demonstrated that CD4^＋T ceils might play important role in production of the pathogenic autoantibodies in pemphigus.
