摘  要:目的探讨邻苯二甲酸二乙基己酯(DEHP)对体外培养的大鼠卵巢颗粒细胞氧化应激和细胞凋亡的影响。方法体外分离和原代培养未成熟大鼠卵巢颗粒细胞,用不同浓度的DEHP(10、50、100nmol/L)染毒24h。应用流式细胞术检测颗粒细胞凋亡率和细胞内活性氧(ROS),应用紫外分光光度计测定颗粒细胞培养液中超氧化物岐化酶(SOD)和丙二醛(MDA)的含量。结果DEHP作用24h后,空白对照组和DEHP低、中、高剂量组细胞凋亡率分别为(2.16±1.14)%、(4.57±1.20)%、(7.72±1.23)%和(12.15±1.29)%;ROS检测结果分别为(4.15±1.18)%、(7.42±1.16)%、(12.09±1.19)%和(19.24±1.15)%;SOD检测结果分别为(17.63±1.21)×10^3、(11.31±1.23)×10^3、(6.89±1.20)×10^3和(3.52±1.22)×10^3U/L;MDA检测结果分别为:(1.23±0.10)×10^3、(2.61±0.11)×10^3、(4.98±0.10)×10^3和(8.21±0.11)×10^3nmo/L。与对照组相比,大鼠卵巢颗粒细胞内ROS和MDA增高,颗粒细胞内SOD酶活性降低,同时颗粒细胞凋亡率增加,呈浓度依赖关系。结论DEHP可通过激活细胞氧化应激而诱导大鼠卵巢颗粒细胞凋亡,进而影响卵巢功能。Objective To investigate the effects of di-2-ethylhexyl phthalate (DEHP) on oxidative stress and apoptosis in mouse ovarian granulosa cells in vitro. Methods Mouse ovarian granulosa cells were separated and primary cultured, and the granulosa cells were treated with different concentrations of DEHP ( 10, 50, 100 nmol/L) for 24 h. Flow cytometry was used to detect apoptosis rate and reactive oxygen species (ROS), and UV-spectrophotometer was used to assess superoxidase dismutasc (SOD) and malonyldialdehyde (MDA) in the cultural medium. Results After treated with DEHP for 24 h, apoptosis rate in control group, DEHP low-dose group, medium-dose group, and high-dose group were (2. 16 ±1.14) %, (4.57 ±1.20) %, (7.72 ±1.23) %, and (12.15 ± 1.29) %, respectively; ROS were (4.15 ± 1.18) %, (7.42±1.16) %, (12.09±1.19) %, and (19.24±1.15) %, respectively; SOD were (17.63±1.21)×10^3, (11.31 ±1.23) ×10^3, (6.89±1.20) ×10^3, and (3.52-±1.22)×10^3 U/L, respectively; and MDA were (1.23 ± 0.10) ×10^3, (2.61 ± 0.11 ) ×10^3, ( 4.98 ± 0. 10)×10^3, and ( 8.21 ± 0.11 ) ×10^3 nmol/L, respectively. Compared with the control group, levels of ROS and MDA in different DEHP groups were increased and levels of SOD were decreased, both in a concentration dependent manner. Meanwhile, apoptosis in different DEHP groups was increased in a concentration dependent manner. Conclusion DEHP could induce apoptosis via oxidative stress in mouse ovarian granulosa cells, resulting in ovarian dysfunction.
