摘  要:在对1例死亡的转基因克隆牛进行病理解剖观察基础上,采用组织学常规石蜡切片-HE染色技术研究克隆牛心脏、肝脏、肺脏和肾脏的组织形态及变化。结果显示,克隆牛体格及脏器体积均偏大。心脏、肝脏出现代偿性增生和淤血,肺脏呈现典型的肺不张和严重局灶性肺气肿。组织学观察显示,心肌纤维拉长变细、横纹不清晰,部分核着色深,出现轻度颗粒变性;肝细胞有明显的脂肪变性、颗粒变性和缺血性病变。肺间质增宽、多个肺泡壁破裂融合,肺间质和肺泡壁毛细血管扩张充血;肾间质毛细血管淤血,近曲小管上皮细胞颗粒变性严重,上皮细胞发生肿胀、部分脱落至管腔。综合病理形态学观察结果显示,该例克隆牛为典型的生后肺不张导致的肺气肿继发其他脏器增生淤血,最后呼吸衰竭而死亡。In the basis of observing the pathological dissections on a transgenic cloned cattle of neonatal death,the histological appearance and changes of heart,liver,lung and the kidney in the cloned cattle were studied by common paraffin sectioned and HE stained methods.The results showed that,both the body and the volume of the main organs of the transgenic cloned cattle were bigger than common cattle,with compensatory hyperplasia and congestion in heart and liver,typical atelectasis and severe focal emphysema in lung.Histological observation showed that,the cardiac muscle fibers were elongated and thinned with unclear transverse striation,some nuclear were stained strongly accompanied by slight granular degeneration.In liver,evident steatosis,granular degeneration and ischemic lesions of hepatocyte could be seen.Interstitial tissue broadened obviously,some alveolar wall were damaged and fused each other.Capillaries both in interstitial tissue and alveolar wall distended and congested seriously.Capillaries in renal interstitium were congested,and typical granular degeneration occurred in epithelial cells of proximal convoluted tubule,some epithelial cells swelled and fell off to lumen.The results above indicated that the cloned cattle mainly died of respiratory failure owing to pulmonary atelectasis leading to emphysema and secondary other organ's proliferation and congestion.
