中药佐剂对免疫幼犬血清抗体效价和细胞因子含量的影响

摘　　要：为了观察中药佐剂对犬疫苗的免疫增强作用,选取45日龄仔犬24头随机分为6组,均用英特威二联苗首免,首免后第14、28天再分别用英特威四联苗、辉瑞四联苗二免、三免,1头份/头,在每次免疫的同时,4个中药佐剂组分别肌肉注射高浓度方Ⅰ和方Ⅱ、低浓度的方Ⅰ和方Ⅱ,1mL/头,佐剂对照组口服左旋咪唑片(2.5mg/kg),无佐剂对照组不给药。分别于免疫前(0d)及首次免疫后第7、14、21、28、35、42天前肢静脉采血,分离血清,检测犬瘟热、犬细小病毒病和犬传染性肝炎抗体及IL-2、IFN-γ、IL-4和IL-10含量的变化。结果显示,2个中药佐剂对3种抗体效价及4种细胞因子的含量均有不同程度的提高,并存在一定的量效关系,综合评价以方Ⅰ高剂量的效果最好。In order to study the immunoenhancement of Chinese herbal medicinal adjuvant,twenty-four 45-old-day puppies were randomly assigned into 6 groups.All puppies were vaccinated with intervet bivalent vaccine,and repeated vaccination with intervet quadruple vaccine and Pfizer quadruple vaccine on days 14 and 28 after the first vaccination respectively.At the same time of every vaccination,puppies in the four Chinese herbal medicine groups were intramuscularly injected 1 mL of prescriptions Ⅰ and Ⅱwith high dose and low dose,respectively,in adjuvant control（AC） group,given Ergamisole 2.5 mg/kg per os,and in non-adjuvant control（NA） group not given any drug.Before vaccination（0 d） and on days 7,14,21,28,35 and 42 d after first vaccination,blood were sampled from forelimb vein for determination of CDV,CPV and CAV antibody,and determination of the content of serum IL-2,IFN-γ,IL-4 and IL-10.The results showed that two Chinese herbal medicinal adjuvants could improve the three kinds of antibody titers and promote the secretion of four kinds of cytokines,and had the dose-effect relationships.In general comparison,the high dose of prescription Ⅰ expressed the best effect.
