摘 要:目的 :用两对特异引物扩增 12株七日热型、澳洲型钩端螺旋体野生株及相应的 2株参考株的 16S、2 3SrRNA基因 ,用Hinf内切酶对扩增产物作MRSP分析。方法 :应用rRNA基因MRSP分析技术。结果 :不同宿主来源 (钩端螺旋体病人、耕牛 )的同一血清型钩端螺旋体野生株之间具有相同的MRSP型 ,同一血清型钩端螺旋体野生株与相应的参考株的MRSP型也相同 ,故七日热型、澳洲型钩端螺旋体属同一MRSP型。结论 :用此法对钩端螺旋体进行快速鉴定 ,初步分类 。To amplified 16S and 23S rRNA genes from 12 field strains of Leptospira interrogans serovar hebdomadis and australis, as well as 2 corresponding reference strains, two pairs of primers were used.The PCR products were digested by endonuclease Hinf I. The results showed: The strains generated identical MRSP patterns as that of the same serovars of Leptospira from different reservoirs (patients with leptospirosis or farm cattles). The field strains and corresponding reference strains produced unique MRSPs providing strains from the same serovars of Leptospira. Leptospira interrogans serovar hebdomadis was classified to the same MRSP patterns with Leptospira interrogans serovar australis. To our viewpoint,this method could be applied to rapidly identify and preliminary classify Leptospira interrogans and one of effective molecular epidemiological means.
