摘 要:目的比较植绒尼龙及棉质取样拭子对大肠杆菌培养物的吸收能力、释放能力及释放均一性。方法 4个稀释度的大肠杆菌培养物,分别用植绒尼龙及棉质取样拭子吸取等量的培养物,进行酶联免疫吸附法(ELISA)检测,比较不同拭子对测定结果的影响。结果以无拭子阳性对照为标准,棉质取样拭子在各个稀释度处吸光度(A)值与阳性对照差异均有统计学意义(P<0.05);而植绒尼龙拭子除在1:40稀释度处有差异,其余各稀释度均与阳性对照差异无统计学意义(P>0.05),明显优于棉质取样拭子;在1:120稀释度上,植绒尼龙拭子与其相应的空白拭子对照差异有统计学意义(P<0.05),而棉质取样拭子与其相应的空白拭子对照差异无统计学意义(P>0.05)。样本释放均一性方面,植绒尼龙拭子的A值变异系数较小,低于棉质取样拭子。吸水性方面,植绒尼龙拭子需3~11s可将大肠杆菌培养物吸收,而棉质取样拭子需15~120s,甚至更长时间。结论植绒尼龙拭子对大肠杆菌培养物的吸收、释放能力强,释放均一性好,优于棉质取样拭子;在ELISA检测临界值附近,棉质取样拭子可能产生假阴性结果。Objective To compare the absorbing ability, release ability and release uniformity of nylon flocked swabs and cotton swabs for detection of colon bacillus culture. Methods Nylon flocked swabs and cotton swabs were used to absorb equivalent colon bacillus culture of 4 different concentrations and detected with ELISA. The impacts of different swabs on detecting results were then compared. Results Based on the standard of no positive swab con- trols, there was significant difference in OD values at every concentration between cotton swabs and positive swab con- trois (P〈0.05). While difference was only found in 1:40 concentration for nylon flocked swabs, the remaining concen- trations were consistent with the positive swab controls (P〉0.05), which was better than that of the cotton swabs. At concentration of 1:120, difference was found between the nylon flocked swabs and their corresponding blank swabs (P〉0.05). For the release uniformity of samples, the variation coefficient of optical density in nylon flocked swabs was relatively smaller than that in cotton swabs. As for water absorbability, nylon flocked swabs just needed 3-11s to ab- sorb the colon bacillus culture, while cotton swabs needed 15 s-2 rain or even longer. Conclusion Nylon flocked swabs had better absorption, release ability and release uniformity as compared with cotton swabs. Around the borderline of ELISA Test, cotton swabs might lead to false negative results.
