摘 要:以牛血清白蛋白为对象 ,研究了丙烯酸 -丙烯酰胺型 (PBuAMA)和疏水改性的四乙烯五胺-环氧氯丙烷型 (HPTE)聚合物凝胶对蛋白质的吸附与释放过程。在吸附过程中 ,当溶液 pH值在牛血清白蛋白等电点 (4.7)附近时 ,凝胶对蛋白质的吸附值最大 ;而在释放过程中 ,pH=7的释放率远大于其它 pH值时的释放率。实验表明凝胶结构对蛋白质吸附与释放有较大影响。未疏水改性的凝胶与疏水改性的凝胶相比 ,前者的释放速率约为后者的2倍。结果表明小分子在凝胶内的扩散以及凝胶与蛋白质的相互作用是影响吸附与释放的重要因素。Poly(acrylic acid-acrylamide-terbutylacrylamide) gels and stearyl hydrophobically modified poly(tetraethylenepentamine-epichlorohydrin) gels were prepared. The absorption and release of protein on polysoap gels were investigated using Bovine Serum Albumin as a model molecule. There are two processes during the absorption: diffuse of protein into the gels and conjugate of protein on the gels. The maximum absorption was observed when the pH of solutions was near the isoelectric point of protein. A larger release ratio was observed at pH 7. The results indicated that the diffusion of protein into the gels and interaction of protein on the gels play an important role during the process of absorption or release. The release ratio of non-hydrophobically modified gels is 2 times faster than hydrophobically modified gels.
