摘 要:目的:验证纤维连接蛋白是否可以作为室间隔缺损(VSD)患者血清标志物。方法:收集室间隔缺损患者血清和与之年龄、性别、民族匹配的房间隔缺损患者、法洛氏四联症患者和健康对照者血清各20例,应用免疫印迹法(Westernblot)和酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测血清样本中纤维连接蛋白表达水平。结果:免疫印迹法检测结果显示,纤维连接蛋白在VSD组、ASD组、TOF组和健康体检者血清中均能检测到,但在VSD组中的表达水平明显高于其他3组(P<0.05);ELISA检测显示VSD组患者血清中纤维连接蛋白浓度为(67.08±27.67)μg/L,ASD组患者为(37.65±15.07)μg/L、TOF组患者为(28.08±11.24)μg/L、健康体检者为(31.15±11.18)μg/L,4组间差异有统计学意义(P<0.05);进一步进行两两比较,VSD组与其他3组差异均有统计学意义(P<0.05),而其他3组差异均无统计学意义(P>0.05)。结论:纤维连接蛋白在VSD患者血清中的含量较高,有可能作为VSD的血清标志物,但有待于进一步的临床大样本验证。Objective: To validate whether fibronectin can be used as a serum molecular marker of patients with ventricular septal defect (VSD) . Methods : The serum samples were obtained from 20 children with VSD, 20 children with atrial septal defect ( ASD), 20 children with tetralogy of Fallot (TOF), and 20 healthy children, who were well matched in age, gender, and race, respectively. Fibronec- tin expression level in serum samples was detected by Western blot and ELISA. Res^dts: Western blot analysis showed that fibronectin was detected in all serum samples of children with VSD, ASD, TOF and healthy children. However, fibronectin expression level in VSD group was significantly higher than that in the other three groups (P 〈 0.05) . The results of ELISA indicated that the serum concentrations of fi- bronectin in VSD group, ASD group, TOF group, and healthy children were (67. 08±27. 67) μg/L, (37. 65 ± 15.07) μg/L, (28.08 ± 11. 24) μg/L, and (31.15 ± 11. 18) μg/L, respectively; there was statistically significant difference among the four groups (P 〈0. 05) ; the results of further comparison between every two groups, there was statistically significant difference between VSD group and the other three groups respectively (P 〈 0. 05), and there was no statistically significant difference among the other three groups ( P 〉 0. 05) . Con- clusion: Fibronectin expression level is higher in serum of patients with VSD, which can be used as a serum biomarker for VSD, but it needs to be further validated in large clinical samples study.
