血清药理实验方法学在抗鼻咽癌中药筛选中的应用

摘　　要：目的探讨血清药理实验方法学在筛选抗鼻咽癌中药方面的作用。方法将SD大鼠80只随机分为10组,实验组(分别灌胃肿节风、黄芪、薏苡仁、千斤藤、黑墨草、养阴清热解毒剂、复方天仙胶囊和鼻咽清毒颗粒)、阳性对照组(灌胃环磷酰胺)、阴性对照组(灌胃生理盐水),动物给药后采血,提取制备含药血清。运用MTT法检测作用药物血清后细胞增殖的改变,并用流式细胞仪(FCM)测定细胞周期的改变。结果肿节风、薏苡仁、复方天仙胶囊和鼻咽清毒颗粒四种中药可抑制CNE1细胞的增殖,其作用具有时间和浓度依赖性;同时可改变细胞周期分布,使多数细胞阻滞于G1期。而黄芪、黑墨草、千斤藤、养阴清热解毒剂无抗鼻咽癌作用。结论血清药理实验方法学是有效地筛选抗鼻咽癌中药的实验方法 ;肿节风、薏苡仁、复方天仙胶囊和鼻咽清毒颗粒是有效的抗鼻咽癌细胞生长的药物,而黄芪、黑墨草、千斤藤、养阴清热解毒剂无效。Objective To investigate the effect of serum pharmacological experiment on the screening of Chinese herbs for nasopharyngeal carcinoma, and establish the effective screening experimental model of TCM resistance of nasopharyngeal carcinoma. Methods SD rats were randomly divided into 10 groups, i.e., experimental, positive control （cytoxan） and negative control （normal control）. After the administration of drug, the serum was collected. MTT assay was used to examine the effect of CTC-DS on the proliferation of CNE1 cell, and flow cytometry was used to observe the cell cycle distribution. Results CTC-DS inhibited the proliferation of CNE1 cell and the effects were in a time-and concentration-dependent manner. CTC-DS could also induce apoptosis while the G1 phase was arrested. Conclusions CTC-DS inhibits proliferation of nasopharyngeal carcinoma cells via induction of apoptosis and arrest of cell cycle.
