摘  要:目的:评价调补肺肾3种方法(补肺健脾、补肺益肾、益气滋肾法)对慢性阻塞性肺疾病(COPD)大鼠JAK/STAT信号转导的影响及远后效应。方法:大鼠随机分为对照组、模型组、补肺健脾组、补肺益肾组、益气滋肾组和氨茶碱组,采用香烟暴露联合反复细菌感染法制备COPD大鼠模型。于第9周对照组、模型组给予生理盐水,其余组分别给予相应药物灌胃,于第20,32周分批取材,观察肺组织病理,p-JAK2,p-STAT1,p-STAT3,p-STAT5蛋白表达及JAK2和SOCS3 mRNA表达。结果:第20,32周时,模型组JAK2 mRNA和p-JAK2,p-STAT1,p-STAT3,p-STAT5蛋白表达较对照组升高(P<0.01),3个中药(补肺健脾方、补肺益肾方、益气滋肾方)组及氨茶碱组较模型组显著降低(P<0.05,P<0.01)。模型组SOCS3 mRNA较对照组升高(P<0.01),3个中药组及氨茶碱组较模型组明显升高(P<0.01),3个中药组明显高于氨茶碱组(P<0.05,P<0.01)。与第20周比较,第32周补肺健脾组JAK2 mRNA和p-JAK2,p-STAT3,p-STAT5蛋白表达均显著降低(P<0.05,P<0.01),补肺益肾组p-STAT3降低(P<0.01),益气滋肾组p-STAT3,p-STAT5降低(P<0.05,P<0.01),氨茶碱组上述指标均无显著性差异。结论:调补肺肾3法可明显减轻COPD肺组织损伤,且具有良好的远后效应,可能与调控JAK/STAT信号转导有关,其中补肺健脾方在降低p-JAK2,p-STAT3,p-STAT5表达方面,补肺益肾方在降低p-STAT3,益气滋肾方在降低p-STAT3,p-STAT5表达方面具有良好的远后效应。Objective: To evaluate the impact and long-term effect of three prescriptions regulating and tonifying lung and kid- ney ( prescription tonifying lung and spleen, prescription tonifying lung and kidney, and prescription tonifying Qi and kidney) on JAK/ STAT signaling of COPD rats. Method: Rats were randomly divided into the control group, the model group, the Bufeijianpi group, the Bufeiyishen group, the Yiqizishen group and the aminophyline group. The COPD rat model was established by smoke inhalations and bacterial infections. In the 9th week, the control group and the model group were administered with normal saline, while the remai- ning groups are orally given corresponding medicines. In the 20th and 32nd week, the rats were sacrificed in batches to observe the pa- thology in their lung tissues, protein expressions of JAK2, STAT1, STAT3, STATS, and expressions of JAK2 and SOCS3 mRNA. Re- sult: In the 20th and 32nd week, protein expressions of JAK2 mRNA and phosphorylation-JAK2, STAT1, STAT3 and STATS in the model group were higher than the control group ( P 〈 0. 01 ), whereas the three traditional Chinese medicine ( TCM ) ( Bufeijianpi, Bufeiyishen and Yiqizishen) groups and the aminophyline group were significantly lower ( P 〈 0. 05, P 〈 0. 01 ). The expression of SOCS3 mRNA in the model group was higher than the control group ( P 〈 0. O1 ) , whereas the level was notably higher in the three TCM groups and the aminophylline group ( P 〈 0. 01 ). The three TCM groups were remarkably higher than the aroinophylline group (P 〈0. 05, P 〈0. 01 ). Compared with the figures in the 20th week, JAK2 mRNA and phosphorylation-JAK2, STAT3 and STATS were significantly lower in the Bnfeijianpi group in the 32nd week ( P 〈 0. 05, P 〈 0. 01 ) , and so did phosphorylation-STAT3 in Bufeiyishen group (P 〈0. 0! ) and phosphorylation-STAT3 and STAT5 in the Yiqizishen group (P 〈0. 05, P 〈0. 01 ).
