摘 要:目的:观察补肾祛寒治尪汤各剂量组对胶原诱导型关节炎(CIA)大鼠的治疗效果及对骨组织和滑膜组织中骨形态发生蛋白-2水平及血管内皮细胞生长因子水平的影响。方法:将74只雄性Wistar大鼠随机分为正常对照组、模型组、甲氨蝶呤组、补肾祛寒治尪汤稀释剂量组(小剂量组)、补肾祛寒治尪汤正常剂量组(中剂量组)和补肾祛寒治尪汤浓缩剂量组(大剂量组),各给药组连续给药28天,观察大鼠的一般情况、关节炎指数积分,荧光实时定量PCR技术测量大鼠膝关节骨组织及滑膜及血管内皮细胞生长因子水平。结果:中剂量组与各组比较关节炎指数积分明显降低,骨形态发生蛋白-2表达水平明显提高,血管内皮细胞生长因子表达水平明显降低(均P<0.05)。结论:补肾祛寒治尪汤治疗胶原诱导型关节炎大鼠有较好疗效,其作用机制可能是因为减少血管内皮细胞生长因子的表达,而减少血管翳形成,减轻了对骨的破坏;同时提高了骨组织及滑膜组织中骨形态发生蛋白-2的表达,从而增强了对骨的修复,促进骨损伤愈合。Objective: To observe the treatment effects of different doses of Bushen Quhan Zhiwang decoction on the rats with collagen-induced arthritis (CIA) and its effects on the levels of bone morphogenetic protein-2 (BMP- 2) and vascular endothelial growth factor (VEGF) in the bone and synovial tissues. Methods:74 male Wistar rats were randomly divided into the normal control group, the model group, the methotrexate group, the thinner-dose group with Bushen Quhan Zhiwang decoction (the low-dose group), the normal-dose group with Bushen Quhan Zhiwang decoction (the medium-dose group), the concentrated-dose group with Bushen Quhan Zhiwang decoction (the high- dose group), each group administered successively for 28 days. Normal condition, arthritis exponential integral, fluorescence real-time quantitative PCR measurement of knee articular bone tissue, synovium BMP-2 and VEGF levels of rats were observed. Results: The arthritis exponential integral of the medium-dose group, compared with other groups, significantly decreased, the BMP-2 mRNA expression level significantly increasing and VEGF mRNA expression level significantly lowering (P 〈 0.05). Conclusion: Bushen Quhan Zhiwang decoction had a good effect in the treatment of CIA rats with its mechanism of reducing the parmus formation and the damage to the bone due to the reduction of the expression of VEGF; while it improved the BMP-2 expression in the bone tissue and synovial tissue, enhancing the restoration and healing of bones.
