HPLC法测定大庆栽培射干中次野鸢尾黄素的含量

摘　　要：目的:测定大庆栽培射干中次野鸢尾黄素的含量。方法:采用高效液相色谱法,色谱柱为INERTSIL ODS-SP柱(4.6 mm×150 mm,5μm),以甲醇-0.2%磷酸水溶液(53:47)为流动相,流速:1.0 mL/min,检测波长为266 nm。结果:次野鸢尾黄素的线性范围为0.047～0.696μg,R2=0.9999,次野鸢尾黄素平均回收率为100.30%,RSD为1.73%(n=5),次野鸢尾黄素的含量分别为0.23%。结论:本实验方法简便、准确,重复性好,可用于射干的质量评价;大庆栽培射干中次野鸢尾黄素的含量大于0.10%,符合《中国药典》要求。Objective：To determine the content of irisflorentin from Belamcanda chinensis in Daqing.Methods：HPLC method was established with column of INERTSIL ODS-SP（4.6mm×150mm,5μm）,mobile phase of methanol-0.2% phosphoric acid solution（53：47）,flow rate of 1.0 mL/min,wave length of 266nm.Results：There was a good linear relationship when irisflorentin was in the range of 0.047～0.696μg（R2=0.9999,n= 5）,average recovery was 100.30%,RSD was 1.73%（n=5） and the content of irisflorentin was 0.23%.Conclusion：This method is convenient,accurate and good in reproducibility.The content of irisflorentin from Belamcanda chinensis in Daqing is more than 0.1%,which accords with the demands of Chinese Pharmacopoeia.
