摘 要:以猪卵泡期和黄体期的不同直径(〈3 mm,3~6mm,〉6mm)卵泡颗粒细胞和卵泡液为研究对象,采用RT-PCR、ELISA技术检测脑源性神经营养因子(BDNF)在不同直径猪卵泡的颗粒细胞和卵泡液中mRNA和蛋白的表达情况。结果表明:BDNF在不同直径的卵泡颗粒细胞和卵泡液中均有表达。各个直径的卵泡颗粒细胞之间表达差异显著,各个直径卵泡液中的含量差异显著。大卵泡颗粒细胞表达量最低,小卵泡液中的含量最低。卵泡期和黄体期卵巢上,中等卵泡中颗粒细胞和卵泡液中BDNF的表达量差异显著。In order to determine the effect of BDNF on reproductive system,expression of BDNF was detected in different sized swine follicle at follicular phase and luteal phase.RT-PCR,ELISA were used to examine the expression of BDNF in the follicular granulosa cells and follicular fluid of pocine.The result shows that BDNF was expressed in every stage of follicular granulosa cells and follicular fluid.The level of BDNF mRNA was significantly different in follicles among different sizes,in which the highest level was found in large follicles.The level of BDNF protein was at high level in follicle of every stage.The concentration of BDNF in medium follicle in luteal phase was higher than that in follicular phase,which is different from that of granulosa cells.
