摘  要:目的观察自体骨膜细胞一藻酸钙凝胶复合体在动物机体中的成骨情况,探讨藻酸钙凝胶与骨膜细胞的最佳配比。方法选择健康成年新西兰白兔96只,按随机数字表法分为6组(A、B、C、D、E组及对照组),每组6只。将取下的双侧骨膜制成0.4ml细胞悬液,并分别取其悬液体积的1,1/4,1/16,1/64,1/256与0.4ml藻酸钙凝胶制备成复合体,植入自体右侧桡骨缺损模型处。于术后2,4,8,12周进行大体观察、X线片检查和组织学观察,同时测定血清碱性磷酸酶(ALP)和血清钙水平。结果细胞数为1/4和1/16的骨膜细胞悬液与藻酸钙凝胶复合体成骨效果最好。结论自体骨膜细胞悬液一藻酸钙凝胶复合体在自体内成骨作用良好,修复骨缺损效果满意,且操作简便,易于临床掌握和应用。Objective To observe bone formation of compound of autologous periosteum cells plus calcium alginate gelatin in animal body and investigate the best mixing ratio of the two components. Methods A total of 96 healthy adult New Zealand rabbits were randomly divided into six groups ( Groups A, B, C, D, E and control group). The excised bilateral periostea from rabbits in each experimental group were all made into 0.4 ml cell suspensions. The compounds were prepared by blending 0.4 ml cal- cium alginate gelatin with 1, 1/4, 1/16, 1/64 and 1/256 of the periosteum cell suspension respectively, and then were applied to autologous right radial defect area. Gross observation, X-ray examination and histological study were carried out at 2, 4, 8, 12 weeks postoperatively. Levels of serum alkaline phos- phatase and serum calcium were determined as well. Results The compounds containing periosteum cell suspension with cell counts of 1/4 or 1/16 and calcium alginate gelatin reached the best osteogenesis. Conclusion Compound of autologous periosteum cell suspension- calcium alginate gelatin induces obvi- ous bone formation and is worthy of clinical application, for it has advantages of satisfactory bone defect repair and easy operation.
