摘 要:目的研究中药补脑止痫散对戊四氮致痫大鼠海马谷氨酸(Glu)和γ-氨基丁酸(GABA)含量的影响,探讨补脑止痫散的抗痫机制。方法 80只大鼠随机分为空白组、模型组、补脑止痫散低、中、高剂量10 d组和低、中、高剂量21 d组。除空白组外,其余各组腹腔注射戊四氮制作癫痫大鼠模型,分别用低、中、高剂量的补脑止痫散灌胃。在预定时间点处死大鼠,取海马切片,用免疫组化法检测海马Glu和GABA的含量变化情况。结果与模型组相比,补脑止痫散中、高剂量组和低剂量21 d组海马神经元Glu含量均显著或极显著地降低,GABA含量均极显著地升高;治疗时间相同而灌胃剂量不同的各组间比较:高剂量组和中剂量组Glu含量均显著低于低剂量组,高剂量组和中剂量组GABA含量均显著高于低剂量组,而中、高剂量组之间比较则均无显著差异;灌胃剂量相同而治疗时间不同的各组间比较:低剂量10 d和21 d组之间有显著差异,中剂量10 d和21 d组、高剂量10 d和21 d组之间无显著差异。结论补脑止痫散能明显降低癫痫大鼠海马Glu含量,提高GABA含量,调节二者之间的平衡;中、高剂量组效果更明显。Objective: To study the effect of Bunaozhixian powder on the content of Glu and GABA in hippoeampus of epileptic rats, and explore its antiepileptic mechanism. Methods: 80 rats were randomly divided into 8 groups: the normal group,the model group,the low,medium and high dosage Bunaozhixian powder groups treated for 10 and 21 days. Kindling epileptic models were induced by PTZ,except the normal group,then the Bunaozhixian powder groups were treated with different dose. All the rats were killed at specific points in time, the content of Glu and GABA in. hippoeampus were examined with immunohistochemistry method. Results: Compared with the model group,except the low dosage group treated for 10 days,the content of Glu in hippocampus of the Bunaozhixian powder groups all reduced significantly ,while the content of GABA all increased significantly;Comparison among groups treated with same course but with different dosage: the content of Glu of the medium and high dosage groups were all significantly lower than the low dosage groups ,while the content of GA- BA of the former were all higher those that of the latter. But there was no significant difference between the medium and high dosage groups;Comparison among groups treated with same dosage but with different course: the content of Glu and GABA of the the low dosage group treated for 21 days are respectively lower and higher significantly than treated for 10 days,while there was no significant difference in the medium and high dosage groups. Conclusion: The Bunaozhixian powder can apparently reduce the content of Glu and increase that of GABA in hippocampus of epileptic rats and regulate the balance between them,meanwhile,the effects of the medium and high dosage group are more obvious than the low dosage group.
