椎间盘内注射转化生长因子-β1诱导椎间盘退变

摘　　要：目的探讨转化生长因子-β1(transforming growth factor-β1,TGF-β1)对正常成年的羊椎间盘组织内环境的影响。方法将TGF-β1注射进羊的椎间盘内,生理盐水和碱性成纤维细胞生长因子(basic fibroblast growing factor,bFGF)分别作为阴性和阳性对照。术前及术后2、4、6个月检查腰椎X线及MRI,观察影像学变化;术后6个月处死动物,取出完整的椎间盘组织,观察椎间盘组织学变化。结果羊L5/L6椎间盘在注射转化生长因子6个月后(与对照组相比较)即出现明显退变。结论外源性TGF-β1可以导致正常椎间盘的退变。Objective To explore the role of transforming growth factor-1 （TGF-β1） in intervertebral disc tissue of normal adult goats. Methods The TGF-β1 was injected into the intervertebral disc of adult goats and saline and basic fibroblast growing factor （bFGF） were used as the negative control and the positive control. Lumbar spinal X-ray and MRI were taken both preoperative and postoperative at 2, 4, 6 months to observe the image changes, and the intervertebral disc tissues were extracted intactly from the goats which were sacrificed after 6 months postoperative to observe the changes of intervertebral disc tissue by histology methods. Results Compared with the control group, L5/L6 intervertebral disc of goats injected with TGF- β1 degenerated significantly after 6 months. Conclusion TGF-β1 can lead to intervertebral disc degeneration.
