摘 要:目的观测大鼠视神经横断伤后视网膜神经节细胞(RGCs)形态学变化及与图形视网膜电流图(P-ERG)波幅值、波潜时变化的关系,同时探讨其对视功能的影响规律。方法采用大鼠球后视神经横断伤动物模型,在伤后不同时间处死动物并取材后,HE染色观察视网膜形态学的动态变化,P-ERG检测视功能状况。结果视神经横断伤后RGCs数目严重下降,2周内RGCs快速减少,以3-9天为明显,2周以后减少缓慢。损伤经过时间与P-ERG-N95波幅值呈负相关,与其峰潜时呈正相关;RGCs数量变化与P-ERG-P50波幅值及波潜时改变密切相关;至伤后4周P-ERG波形近乎消失。结论P-ERG起源于RGCs,RGCs进行性丧失是P-ERG变化的重要病理基础,并与视功能的时间规律变化具有相关性。Objective To observe the morphological changes of retina and the relationship with the amplitude and the latency of P-ERG in rats, meanwhile, to investigate its effects on visual function after optic nerve transection. Methods In this experiment, intraorbital optic nerve transection model of rats was used. paraffin section was cut after the test group rats were killed after various survival interval, the hemotoxylin and eosin (HE) staining was carried out to observe the morphological changes of retina and the visual function was recorded in normal and injured rats by P-ERG. Results The number of Retinal ganglion cells (RGCs) were reduced significantly due to optic nerve transection(ONT). An initial loss was accelerated by 2 weeks after ONT, RGCs lossed fastly and abruptly during 3 to 9 days after optic nerve transection, the later declines was protracted in RC, Cs densities. There was a negative correlation between post-injury interval and the amplitude of P-ERG- Ps0 wave and a positive correlation between post-injury interval and the latency of P-ERG- Ps0 wave. P-ERG waveform had almost vanished by 4 weeks after optic nerve transection. Conclusion the source of P-ERG is the retinal ganglion cells, the progressing loss of RGCs following optic nerve transection is the important morphological foundation of the changes of P-ERG, which have relativity with the regular decline of visual function of retina.
