摘  要:目的:建立川黄柏、关黄柏中6种有效成分含量的同时测定方法。方法:以HPLC采用Diamonsil C18色谱柱(4.6mm×250 mm,5μm),乙腈-水(1‰乙酸,2mmol醋酸铵)为流动相梯度洗脱,检测波长280 nm,柱温25℃,流速1 mL.min-1。结果:绿原酸、黄柏碱、木兰花碱、药根碱、巴马汀和小檗碱分别在20.00~320.00,18.75~130.00,25.00~200.00,5.00~100.00,20.00~200.00,0.09~1.80 mg.L-1峰面积和质量浓度线性关系良好(r≥0.999 0),平均加样回收率分别为98.1%,99.4%,97.5%,97.3%,104.0%,98.5%,RSD分别为0.5%,0.6%,0.8%,1.0%,1.4%,0.9%。结论:含量测定方法简便、稳定、重复性好,为黄柏药材质量控制和评价提供了依据。Objective: To establish the method for simultaneous determination of 6 active components. Method: Simultaneous determination of chlorogenic acid, phellodendrine, magnoflorine, jatrorrhizine, palmatine and berberine in Huangbai was carried out by HPLC. A Diamonsil Cls (4.6 mm× 250 mm, 5μm) column was used. The mobile phase was aeetonitrile-water (1%o acetic acid, 2 mmol ammonium acetate) solution in gradient elution. The detection wavelength was set at 280 nm, the column temperature was kept at 25℃ and the flow rate was 1 mL·min-1 Result: The linear-range of chlorogenic acid, phellodendrine, magnoflorine, jatrorrhizine, palmatine and berberine was 20.00-320.00, 18.75-130.00, 25.00-200.00, 5.00-100.00, 20.00-200.00, 0.09-1.80 mg·L-1 separately. The average recovery was 98.1% , 99.4% , 97.5% , 97.3% , 104.0% , 98.5% separately; and RSD was 0.5% , 0.6% , 0.8% , 1.0% , 1.4% , 0.9% separately. Conclusion: The method is convenient, stable, reliable and suitable for quality control of Huangbai.
