摘 要:目的:比较人血清透明质酸(HA)夹心化学发光免疫分析和竞争化学发光免疫分析,对两种方法的相关性、线性范围和检测效果进行评价。方法:分别应用人血清HA夹心和竞争化学发光免疫分析对197例血清样本进行HA含量检测,并对检测结果进行分析,同时对两种方法的精密度、回收率和灵敏度进行比较。结果:夹心法的准确性和精密度都要高于竞争法,在95%可信限下其检测灵敏度能达到0.66μg/L,而竞争法只能达到7.26μg/L,两种方法均能准确测定血清中HA含量,其相关性良好,线性关系为y=1.1893x-25.974(r=0.9871),两组数据之间没有显著性差异(t=1.966,P>0.05)。结论:两种方法都能准确测量人血清中HA含量,但夹心化学发光免疫分析的灵敏度、重复性、回收率都优于竞争法。Objective To evaluate the correlation, linear range and detection effect of sandwich and competitive chemilumines- cence immunoassay for the detection of hyaluronic acid from human serum, the performance of the 2 methods was compared. Methods The level of hyaluronic acid in 197 sera was detected by the sandwich and competitive chemiluminescence immunoassay, and the re- sults were analyzed statistically. The precision, recovery and sensitivity of the 2 methods were assessed simueltaneously. Results The accuracy and precision of sandwich chemiluminescence immunoassay were better than competitive chemiluminescence immunoassay. The detection limit of sandwich chemiluminescence immunoassay could reach 0.66 Izg/L, and the competitive chemiluminescence im- munoassay just could reach 7.26 ixg/L. However, both methods could exactly detect hyaluronic acid from serum. The correlation of them was y = 1.1893x - 25. 974 ( r = 0.9871 ), whose results showed no significant difference ( t = 1. 966, P = 0. 664). Conclusion Sandwich and competitive chemiluminescence immunoassay could all exactly detecting hyaluronic acid concentration of serum, but sensitivity, repeatability and recovery of sandwich method were better than those of competitive method's.
