摘  要:目的评价海马神经元γ-氨基丁酸A(GABAA)受体在异氟醚导致新生大鼠认知功能障碍中的作用。方法出生7dSD大鼠28只,雌雄不拘,体重16~18g,采用随机数字表法,将大鼠随机分为4组(n=7):对照组(Con组)腹腔注射生理盐水0.2ml;一叶款碱组(Sec组)于相应时点腹腔注射GABA。受体特异性拮抗剂一叶蔌碱30mg/kg;异氟醚组(Iso组)腹腔注射生理盐水0.2ml,持续吸入1.4%异氟醚6h;一叶蔌碱+异氟醚组(Sec+Iso组)吸人异氟醚前30min和吸入3h时腹腔注射一叶蔌碱30mg/kg,持续吸入1.4%异氟醚6h。给药后3周时处死大鼠取海马制作海马切片,行电生理实验,每隔5min测定群峰电位振幅(PSA)和长时程增强(LTP),记录LTP诱发成功情况。结果与Con组比较,Is0组各时点PSA和LTP诱发成功率降低(P〈0.01),Sec组和Sec+Iso组上述指标差异无统计学意义(P〉0.05);与Is0组比较,Sec+Iso组各时点PSA和LTP诱发成功率升高(P〈0.01)。结论GABA。受体参与了异氟醚导致新生大鼠认知功能障碍的过程。Objective To evaluate the role of gamma-aminobutyric acid A (GABAg ) receptor in isoflu- rane-induced cognitive dysfunction in neonatal rats. Methods Twenty-eight 7-day-old Sprague Dawley rats, weighing 16-18 g, were randomly divided into 4 groups (n = 7 each): control group (group Con), securinine group (group Sec), isoflurane group (group Iso) and seeurinine + isoflurane group (group See + Iso) . Normal saline 0.2 ml was injected intraperitoneally in group Con. Specific GABAA receptor antagonist securinine 30 mg/kg was injected intraperitoneally at the corresponding time points in group Sec. Normal saline 0.2 ml was injected in- traperitoneally and 1.4% isoflurane was inhaled for 6 h in group Iso. Seeurinine 30 mg/kg was injected intraperito- neally at 30 min before isoflurane inhalation and 3 h of inhalation, and 1.4% isoflurane was inhaled for 6 h in group Sec + Iso. The rats were then sacrificed at 3 weeks after administration, their brains were immediately re- moved and hippocampal slices were prepared for electrophysiological experiments. The value of population spike amplitude (PSA) and long-term potentiation (LTP) were measured every 5 min. The successful LTP induction was recorded. Results Compared with group Con, the values of PSA and rates of successful LTP induction were sig- nificantly decreased in group Iso (P 〈 0.01 ), and no significant change was found in the parameters mentioned above in groups See and Sec + Iso ( P 〉 0.05) . The values of PSA and rates of successful LTP induction were sig- nificantly higher in group Sec + Iso than in group Iso ( P 〈 0.01 ). Conclusion GABAA receptor is involved in isoflurane-induced cognitive dysfunction in the neonatal rats.
