摘  要:目的通过纳米金对HepG2上清液诱导的人淋巴细胞进行干预,初步观察纳米金对淋巴细胞增殖活性的影响。方法建立HepG2上清液与人淋巴细胞共培养体系,实验分为对照组、共培养组、纳米金干预组。噻唑蓝(MTT)比色法检测淋巴细胞增殖抑制率;ELISA法检测共培养体系中血管内皮生长因子(VEGF)水平;原子力显微镜(AFM)表征淋巴细胞表面形貌及超微结构。结果 HepG2上清液(富含VEGF)明显抑制淋巴细胞增殖,且随浓度的增大其增殖抑制率上升到(41.90±1.32)%,而纳米金干预后抑制率下降为(35.73±1.54)%(P<0.05);AFM结果显示HepG2上清液诱导的淋巴细胞高度、粗糙度均减小,细胞膜内陷,而纳米金干预组这种变化不明显;共培养组体系中VEGF含量为(308.51±21.73)pg/mL,而纳米金干预组VEGF下降为(96.78±11.27)pg/mL(P<0.05)。结论纳米金通过与HepG2上清液中VEGF结合,增加了共培养体系中淋巴细胞的增殖活性。【Objective】 To observe the effects of gold nanoparticles (GNP) on the proliferation activity of human lymphocyte induced by hepatocellular carcinoma cell (HepG2) supernatants. 【Methods】 Establish coculture system of human lymphocyte and HepG2 cell supernatants, then divide into control group, co-cultured group, and GNP intervention group. The inhibition effect of lymphocyte was assessed by MTT assay; the levels of VEGF in co -culture system was detected by ELISA; then observed the morphology and ultratructure of lymphocyte membrane surface by atomic force microscope (AFM). 【Results】 The HepG2 supernatants is rich with VEGF, which can inhibit the proliferation of lymphocyte significantly. The inhibition rate rose to (41.90± 1.32)% with increasing the concentration of HepG2 supernatants while it reduced to (35.73±1.54)% after intervened by GNP, P 0.05; AFM results show that compared with control group, the changes in morphology and ultratructure of lymphocyte in co-culture group were more obvious, they appeared with invagination membrane, the height and roughness were decreased, while the changes in GNP intervention group were less obvious; the level of VEGF in co-culture group (308.51 ± 21.73 pg/mL) was much more than GNP intervention group (96.78±11.27 pg/mL), P 0.05. 【Conclusion】 Gold nanoparticles can combine with VEGF in HepG2 supernatants, then increase the proliferation activity of lymphocytes in this co-culture system.
