摘 要:目的:采用高效液相色谱法同时测定复方甘草片中吗啡、磷酸可待因、甘草苷和甘草酸的含量。方法:采用PhenomenexLuna-C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.02 mol·L-1磷酸二氢钾溶液(磷酸调pH至4.0)为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长为220 nm(0~23 min,检测吗啡和可待因)、254 nm(23~50 min,检测甘草苷和甘草酸),柱温为35℃。结果:4个成分的峰面积与浓度的线性关系良好(r>0.9999);加样回收率为98.0%~100.8%。结论:该方法可用于复方甘草片中吗啡、磷酸可待因、甘草苷和甘草酸4个成分的含量测定。Objective:To develop an HPLC method for content determination of morphine, codeine phosphate, liquiritin, glycyrrhizic acid in compound liquorice tablets. Methods:The HPLC analysis was performed on a Phe- nomenex Luna C 18 (250mm×4.6mm,5μm) column. The mobile phase consisted of acetonitrile and 0.02 mol . L- 1 potassium dihydrogen phosphate ( pH was adjusted to 4. 0 by phosphoric acid) with a gradient elution at the flow rate of 1.0 mL . min -1. The detected wavelengths were set separately in different time period (0 - 23 min,220 nm for morphine and codeine ;23 -50min,254 nm for liquiritin and glycyrrhizic acid), and the column temperature was 35 ℃. Results:Four active components were separated completely in 50 minutes, and had good linear relation- ship (r 〉 0. 9999 ) and recoveries(98.0% - 100. 8% ) in their respective detection scopes. Conclusion :The method can be applied for simultaneous determination of four active components ( morphine, codeine phosphate, liquiritin, glycyrrhizic acid) in compound liquorice tablets.
