摘 要:目的探讨神经营养因子3(NT-3)与神经干细胞(NSCs)联合移植治疗乳鼠缺血缺氧性脑损伤(HIBD)后的神经电生理现象。方法 10只7日龄Wistar乳鼠随机分配到对照组,50只7日龄Wistar乳鼠制备HIBD模型,存活37只,将其随机分配到NSCs移植组、NT-3联合NSCs移植组、脑损伤组。3 d后NT-3联合NSCs移植组移植NSCs及NT-3,NSCs移植组只移植NSCs,脑损伤组移植PBS。移植后3周、7周时进行神经电生理检测,然后取脑组织行HE染色。结果神经电生理:与脑损伤组相比,移植后3周时,NSCs移植组、NT-3联合NSCs移植组乳鼠的后肢股四头肌复合动作电位(CMAP)波幅明显增高、潜伏期缩短,以NT-3联合NSCs移植组为著(P均<0.05);移植后7周时,CMAP波幅明显增高(P<0.05),潜伏期缩短不明显;移植后3、7周时HE染色:脑损伤组损伤侧侧脑室旁正常组织结构破坏,出现多个大小不一的囊腔;NSCs移植组侧脑室旁正常组织结构有所恢复,囊变少见;NT-3联合NSCs移植组损伤侧侧脑室周围组织结构恢复更好。结论 NT-3联合NSCs移植可使HIBD乳鼠后肢股四头肌CMAP波幅明显增高、潜伏期缩短。Objective To observe neural electrophysiological manifestations of neonatarats with hypoxia ischemia brain damage(HIBD) in the treatment of neurotrophin-3 (NT-3) and neural stem cells (NSCs) transplantation. Methods Totally 10 7-day-old Wistar neonatal rats were randomly assigned to the control group, 50 7-day-old Wistar neonatal rats were selected to establish HIBD animal models, 37 cases were alive, and then randomly assigned to the NSCs transplanta- tion group, NT-3 jointed NSCs transplantation group, brain injury group. NSCs, NSCs + NF3, PBS were transplanted in the injury side of the lateral ventricle respectively after 3 days. After transplanted for three weeks, seven weeks, the nerve electrophysiological was tested, then taking the brain tissue for HE staining. Results Neurophysiological testing: after 3 weeks for transplantation, compound action potential (CMAP) in rat hind limb quadriceps of rats with NT-3 combined NSCs transplantation or NSCs transplantation was significantly higher volatility, shorter incubation period compared with brain injury group( all P 〈 0.05) ; after 7 weeks for transplantation, CMAP in rat hind limb quadriceps of rats with NT-3 combined NSCs transplantation was significantly higher volatility(P 〈 0.05 ) ; HE staining: in brain injury group, damage lateral ventricle adjacent normal tissue structural damaged, there were a number of different sizes of the cysts, in NSCs transplantation group, periventricular normal organizational structure had been restored, and had rare cystic; in NT-3 joint- ed NSCs transplantation group, the ipsilateral periventricular tissue structure recovered more better. Conclusions NT-3 combined neural stem cell transplantation can make CAMP amplitude in hind limb quadriceps of HIBD rats increased, the incubation period shorted.
