摘  要:目的探讨慢性肾衰竭肾组织骨形成蛋白-7(BMP-7)和转化生长因子β1(TGF-β1)表达水平及药物干预的作用。方法 SD大鼠5/6肾切除法构建慢性肾衰竭模型,荧光定量聚合酶链反应测定肾组织TGF-β1及BMP-7表达,观察各研究组肾组织TGF-β1及BMP-7表达情况及血尿素氮(BUN)、肌酐(Cr)及尿蛋白水平。结果与对照组比较,未处理组TGF-β1水平增高、BMP-7水平降低(P<0.05)。洛汀新组TGF-β1、BMP-7水平低于对照组(P<0.05),且BMP-7水平低于未处理组。肾衰康组TGF-β1水平低于病理组(P<0.05),BMP-7水平高于未处理组(P<0.05);洛汀新组及肾衰康组血BUN、Cr及尿蛋白水平均低于未处理组(P<0.05)。结论慢性肾衰竭肾组织TGF-β1表达增强、BMP-7表达降低;洛汀新可抑制TGF-β1表达,但无增强BMP-7表达的作用;肾衰康既能抑制TGF-β1表达,也能增强BMP-7表达。Objective To evaluate the expression of bone morphogenetic protein-7(BMP-7) and transforming growth factor-β1(TGF-β1) in renal tissues of chronic renal failure and the efficacy of drug intervention.Methods Chronic renal failure models were established by nephrectomize 5/6 kidney of SD rats,and the expression of BMP-7 and TGF-β1 of kidney were detected by fluorescence quantitative polymerase chain reaction,and the concentration of blood urea nitrogen(BUN),creatinine(Cr) and urine protein of different groups were observed.Results The expression of TGF-β1 was significantly increased and BMP-7 decreased in untreated group compared with control groups(P0.05).The expression of TGF-β1 was significantly decreased in Lotensin group,compared with control group(P0.05),but the expression of BMP-7 was decreased,compared with control and untreated groups(P0.05).The expression of TGF-β1 was significantly decreased in Lotensin group,compared with untreated group,otherwise,the expression of BMP-7 was opposite(P0.05).The concentration of BUN,Cr and urine protein were significantly decreased in Lotensin group and Shenshuaikang group,compared with untreated group(P0.05).Conclusion The expression of TGF-β1 might be increased and BMP-7 decreased in chronic renal failure.Lotensin could inhibit the expression of TGF-β1,but could not enhance the expression of BMP-7.Shenshuaikang could inhibit the expression of TGF-β1 and enhancing the expression of BMP-7.
