孟鲁司特对哮喘小鼠肺组织黏蛋白Muc5ac表达的影响

摘　　要：目的研究白三烯受体拮抗药孟鲁司特对哮喘小鼠肺组织中黏蛋白Muc5ac表达及支气管肺泡灌洗液中白细胞介素13(IL-13)分泌的影响,探讨孟鲁司特对哮喘气道炎症黏液高分泌影响的机制。方法 18只清洁级BALB/c小鼠随机分为哮喘组、孟鲁司特组和正常对照组,每组6只。免疫组化法检测Muc5ac蛋白在小鼠肺组织的表达。收集支气管肺泡灌洗液(BALF),酶联免疫吸附测定(ELISA)法检测其中IL-13的水平。结果 Muc5ac蛋白在哮喘小鼠肺组织中的表达为(0.572±0.028),较正常对照组(0.137±0.041)明显升高,孟鲁司特治疗组Muc5ac蛋白表达(0.497±0.054)较哮喘组明显降低。哮喘组的BALF中IL-13为(78.07±6.78)pg.mL-1,较正常对照组(28.58±12.98)pg.mL-1明显升高,孟鲁司特组为(51.2±3.53)pg.mL-1,较哮喘组明显降低。Muc5ac蛋白表达与BALF中IL-13水平呈直线正相关(r=0.861,P<0.05)。结论孟鲁司特可能通过下调IL-13和Muc5ac蛋白的表达,对哮喘气道黏液高分泌发挥治疗作用。Objective To study effects of leukotriene receptor antagonist montelukast on the expression of mucoprotein MucSac in lung and IL-13 secretion in bronchoalveolar lavage fluid （BALF） of asthmatic mice and investigate the possible mechanism. Methods Eighteen BALB/c mice were randomly divided into the control, asthmatic model and montelukast groups, 6 mice in each group. The expressions of Muc5ac protein in lung were detected by immunohistochemical assay. Interleukin-β level in BALF were analyzed via ELISA. Results The MucSac expression in asthmatic group （0. 572 ± 0. 028 ） was significantly upregnlated compared with control group （0. 137±0. 041, P 〈0. 01 ）. MucSac protein in the montelukast treated group （0. 497± 0. 054） was evidently lower than that of the asthmatic group（P〈0.05 ）. The level of IL-13 in BALF in the asthmatic group（78.07± 6.78） pg . mL^-1 was obviously increased compared with the control group（28.58±12.98） pg . mL^-1 ,P〈0.01, while, which in the montelukast group （51.2±3.53） pg . mL^-1 was distinctly lower than that of the asthmatic group （P〈0.01）. MucSac protein expression was positively correlated to the level of IL-β （r = 0.861,P〈0.05）. Conclusion Montelukast plays a therapeutic role in mucus hypersecretion of asthmatic airway, which is associated with down-regnlation of MucSac and IL-β.
