摘  要:目的筛选大孔吸附树脂纯化藤梨根中总黄酮的工艺条件。方法通过动态吸附和解吸的方法,以总黄酮的吸附率、解吸率为评价指标确定树脂型号;通过最佳上样量、上样速度、水洗用量、乙醇洗脱流速、乙醇解析浓度选择、乙醇解析用量等的筛选实验,确定纯化工艺条件。结果 AB-8型大孔吸附树脂分离纯化藤梨根中总黄酮效果最好,其吸附率为84.76%,解析率为95.51%。其最佳工艺条件为:30 mL藤梨根提取液以3 BV.h-1的流速通过树脂柱,先用水8 BV洗涤,洗脱流速为4 BV.h-1,再用70%乙醇4 BV解析,洗脱流速为3 BV.h-1。结论 AB-8型大孔树脂在所确定的优化工艺条件下,可较好地吸附分离藤梨根中总黄酮。Objective To study the purification process for total flavonoids from Tengligen with macroporous adsorption resin. Methods Dynamic adsorption-desorption was adopted to screen the optimal type of resin by measuring the adsorption ratio, eluting ratio of total flavonoids. The purification procedure was confirmed via determination of optimal loading volume and velocity, water-eluting amount, eluting rate, desorption concentration and volume of ethanol. Results The AB-8 macroporous adsorption resin presented the best separating efficiency, the rate of adsorption was 84. 76%, the rate of desorption was 95.51%, loading 30 mL extracts, flowing at 3 BV·h^-1 , with eluting agent as 8 BV purity water eluting at 4 BV·h^-1 , and 4 BV 70% ethanol, and eluting at 3BV·h^-1. Conclusion Total flavonoids from Tengligen can be well adsorbed and separated under the optimized conditions with AB-8 macroporous absorbent resin.
