摘 要:目的观察周期性单轴牵张应力对大鼠前软骨干细胞增殖相关基因的影响。方法采用四点弯曲应力仪分别以2 000μstrain和4 000μstrain的牵张力刺激前软骨干细胞1 h,并设空白对照组,荧光定量PCR检测相关增殖基因PCNA和Cyclin D1的表达情况,并用免疫荧光检测力学激前后Ki67阳性细胞的表达情况。结果 2 000μstrain刺激组细胞表达PCNA以及CyclinD1较对照组高(P<0.05),免疫荧光检测提示2 000μstrain刺激组的细胞核内Ki67表达较对照组明显增多;而4 000μstrain刺激组细胞PCNA的表达与对照组相比明显降低(P<0.05),而Cyclin D1降低不明显(P>0.05)。结论适宜的周期性单轴牵张力能引起大鼠前软骨干细胞相关增殖基因的表达增加。Objective To investigate the impact of cyclical tension force on the proliferation of precartilagious stem cells (PSCs). Methods PSCs were stimulated with 2 000 and 4 000μstrain for 1 h. The expression of PCNA and Cyclin D1 in each group was examined by using PCR. The expression of Ki67 was detected by using immunofluorescent staining. Results The mRNA expression of PCNA and Cyclin D1 in 2 000 μstrain group was increased significantly as compared with the control group, and the expression of PCNA in 4 000μstrain group was reduced as compared with control group, but the expression of Cyclin D1 was not reduced obviously. Immunofluorescent staining revealed the expression of Ki67 in 2 000 μstrain group was increased significantly as compared with control group. Conclusion Proper cyclic tensile strain could increase the expression of PSCs proliferation-related genes in rats.
