摘 要:目的:通过液相色谱-串联质谱(LC-MS/MS),采用选择监测离子法测定补虚颗粒中升麻素苷的含量。方法:样品用60%甲醇提取,通过液相色谱-串联质谱测定其中升麻素苷的含量,在正离子模式下,采用选择离子监测(SIM)方式进行测定,其监测离子对为m/z469→307。结果:升麻素苷在1~100μg.mL-1范围内线性关系良好(r=0.998 3),平均回收率96.0%,检测限为1.0ng.mL-1。结论:该方法快速、灵敏,结果准确,适用于复杂中药基质中升麻素苷的检测。OBJECTIVE To establish a rapid and sensitive method for the determination of prim-(Yglucosylcimifugin (POG) in Buxu granule by LC-MS/MS using selection ion monitoring (SIM). METHODS The sample was extracted by 60% aqueous methanol. The mass spectrometer was operated in the positive ionization electrospray (ESI) mode using SIM. The lransitions m/z 469→3117 was used to quantify POG. RESULTS The method had satisfactory linearity in the range of 1 - 101) μg.mL-1 with a regression coefficient of (0. 998 3, the limit of detection (LOD) was 1.0 ng.mL-1 , and the mean recovery was 96%. CONCLUSION The method is sensitive, simple and accurate, and proved to be suitable in the determination of POG in the com- plicated traditional Chinese medicine matrices.
