摘 要:目的通过研究"和血生络方"对心肌梗死模型大鼠左室心肌组织AKT mRNA表达的影响,从信号转导通路角度探讨"和血生络法"促血管新生的作用机制。方法选雄性Wistar大鼠,制作心肌梗死大鼠模型,随机分为消心痛组,和血生络高、低剂量组及模型组,同时建立假手术组,每组6只。药物治疗组分别给予和血生络水溶液(9.9,19.8 g.kg-1.d-1)及消心痛水溶液(2.7 mg.kg-1.d-1)每天灌胃1次,其他组给予等量蒸馏水,连续用药3 w后处死大鼠,取心脏组织检测AKT mRNA的表达。结果模型组较假手术组大鼠显著增高(P<0.05)。和血生络高、低剂量组及消心痛组明显高于模型组(P<0.05)。和血生络高剂量组与消心痛组明显优于和血生络高低剂量组(P<0.05)。和血生络高剂量组明显优于消心痛组(P<0.05)。结论 PI3K/Akt信号通路在"和血生络法"促血管新生中起着重要作用。Objective To observe the expression of AKT mRNA in left ventricular myocardial tissue in rats and discuss angiogenesis promoting mechanism of Hexue Shengluo method from the signal transduction pathways. Methods Myocardium of infarcted rats model were made by Wistar rats. They were divided randomly into model, Y, G and D groups. Rats in sham group were made at the same time. Every group had 6 rats and rats were lavaged after 24 hours. Rats in drug treatment groups were administrated with ID water solution in accordance with 2. 7 mg·kg^-1·-d^-1 and with Hexue Shengluo water solution in accordance with 9. 9 and 19. 8 g·kg^-1·-d^-1. Rats in model and sham groups were treated with distilled water with the equal volume. Rats were treated once every day by intragastric administration. After 3 weeks continuously, rats were executed and hearts were got. Expression of AKT mRNA of cardiac tissue was detected. Results The expression of AKT mRNA in model group was higher than that of sham group ( P 〈 0. 05). The expression of AKT mRNA in therapeutic groups were higher than that of model group (P 〈 0. 05 ). High dose of Hexue Shengluo and isosorbide dinitrate group were higher than that of low dose of Hexue Shengluo group ( P 〈 0. 05 ). High dose of Hexue Shengluo group was better than that of isosorbide dinitrate group (P 〈 0. 05 ). Conclusions PI3K/Akt signal pathway may play an important role in Hexue Shengluo promoting angiogenesis.
