摘  要:采用阶段诱导法进行体外诱导肝上皮样干细胞(WB—F344)为胰岛素分泌细胞(IPCs),将IPCs移植至糖尿病大鼠体内,监测血糖变化。结果显示,RT—PCR和免疫荧光技术分别检测到IPCs中表达胰岛β细胞的特异性转录因子及蛋白。IPCs移植糖尿病大鼠体内后,明显改善大鼠的高血糖状态。提示肝上皮样干细胞可以在体外经小分子重编程为有功能的胰岛素分泌细胞,其有助于糖尿病的治疗。Using phase induction in vitro to induce hepatic epithelial-like stem cells ( WB-F344 ) into insulin producing cells ( IPCs ) , the IPCs were transplanted into diabetic rats, and the blood glucose level was monitored. The results of RT-PCR and immunofluorescence showed that genes and proteins of pancreatic islet beta cell and phenotypic markers were successfully detected in the IPCs, which significantly improved hyperglycemia after being transplantated into diabetic rats. These results suggest that hepatic epithelial-like stem cells in vitro can be induced into functional insulin producing cells by small molecules and that this finding may contribute to the treatment of diabetes.
