摘 要:目的制备丹参酮ⅡA纳米囊,并评价其性能。方法采用乳化-溶剂挥发法制备丹参酮ⅡA纳米囊,以包封率为指标,单因素试验结合正交试验优化制备工艺与处方,并评价纳米囊的粒径形态。结果以乙酸乙酯-水(1∶2)、0.5 mL司盘-80、丹参酮ⅡA-壳聚糖(1∶5)、乙醇0.4 mL为最佳处方,在温度为50℃,搅拌速度为300 r/min条件下乳化,然后超声30 s,即得纳米囊。所得纳米囊外观圆整,平均粒径(172±10)nm,包封率高达73.2%。结论乳化-溶剂挥发法适用于制备丹参酮ⅡA纳米囊,且工艺稳定。Objective To prepare the tanshinone ⅡA nanocapsules and to evaluate its quality. Methods Tanshinone ⅡA nanocapsules were prepared by emulsion solvent diffusion method and were optimized by both of single factor test and orthogonal design with encapsulation ratio as index. The shape and diameter of tanshinoneⅡA nanocapsules were investigated. Results The optimal formulas were obtained under the following conditions: the recipe consisting of ethyl acetate-water(1 : 2), 0.5 mL of span-80, Tan ⅡA -chitosan(1 : 5), and 0.4 mL of ethanol, emulsification after stirring in the speed of 300r/min at 50℃, and ultrasonic time being 30 s. Under the optimal conditions, the obtained tanshinone ⅡA nanoeapsules were uniform and round, and had the average diameter of (172 ± 10) nm, the entrap- ment ratio reached 73.2 %. Conclusion Emulsion solvent diffusion method is applicable for the preparation of tanshi- none ⅡA nanocapsules, the process being steady.
