摘 要:目的:探究海参二十碳五烯酸磷脂(Eicosapentaenoic Acid Phosphatidylcholine,EPA-PC)对链尿佐菌素(Streptozocin,STZ)诱导的糖尿病大鼠肾脏的保护作用机制。方法:采用两次腹腔注射STZ的方法建立糖尿病大鼠模型,经不同剂量的海参EPA-PC(25、75 mg/kg,以EPA含量计)灌胃8周后,分别检测大鼠空腹血糖、葡萄糖耐量以及肾肥大指数;光镜观察肾组织的显微结构变化;采用聚合酶链反应(Reverse Transcription PolymeraseChain Reaction,RT-PCR)技术检测肾皮质中转化生长因子(Transforming Growth Factor-β1,TGF-β1)及其Ⅱ型受体(Transforming Growth Factor-β1 Receptor,TβRⅡ)、结缔组织生长因子(Connective Tissue Growth Factor,CTGF)和过氧化酶体增殖物活化受体γ(Peroxisome Proliferator Activated Receptorγ,PPARγ)mRNA的表达水平。结果:海参EPA-PC能显著地改善糖尿病大鼠的葡萄糖耐量和肾肥大指数(P<0.05、P<0.01);有效修复肾小囊肿胀,抑制肾小球肥大;显著地下调TβRⅡ、TGF-β1和CTGF mRNA的表达(P<0.01),同时上调PPARγmRNA的表达(P<0.01)。结论:海参EPA-PC对糖尿病大鼠肾脏具有较好的保护作用,其作用机制与下调糖尿病大鼠肾脏中TβRⅡ、TGF-β1、CTGF,及上调PPARγmRNA的表达有关。[Objective] To study the protective mechanism of eicosapentaenoic acid phosphatidylcholine (EPA-PC) on the renal injury of diabetic rats induced by STZ. [Method]Diabetic rats were induced by STZ twice, and 25 and 75 mg/ (kg · bw) of EPA-PC were given for 8 weeks respectively. At the end of the experiment, the level of fasting blood glucose, oral glucose tolerance and renal index were determined. Microstructure of the kidney was observed, then mRNA expression of TGF-β1, TβRII,CTGF and PPARγ in the kidney were assessed by reverse transcription polymerase chain reaction (RT-PCR) . [Result ]EPA-PC had remarkably decreased the level of oral glucose tolerance and renal index, reverted the microstructure of renal lesion, and markedly suppressed the mRNA expressions of TGF -β1, TβR lI, CTGF mR- NA, while upregulated the mRNA expression of PPART ( P 〈 0. 01 ) . [Conclusion] It suggested that the markedly protective effects of EPAPC on kidney might related to inhabiting the mRNA exppertion of TβR 11, TGF -β1, CTGF and upregulating PPARy.
